Objective: To observe the effects of sclera apoptosis on the form deprivation myopia of Nourishing blood and Tonifying kidney decoction. Methods: The models of form deprivation myopia were made through translucent goggles monocular covered 60 d with two weeks healthy guinea pigs. Building successfully guinea pigs were randomly divided into two groups, control group and dosing group. The diopter and eye axis were measured before and fater covered. The guinea pigs of dosing group were continuously lavaged 15 d with Nourishing blood and Tonifying kidney decoction, one time a day. To observe the morphological changes of sclera organization by HE staining. The control group had no intervention. To compare the difference between covered eyes and control eyes. To observe the cell apoptosis condition of sclera cells by using terminal deoxynucl neotidyl transferase mediated d UTP nick end labelling method and flow cytometry. And to observe the sclera ultrastructure of both groups by transmission electron microscopy technology. Results: The models of form deprivation myopia were made successfully. The diopter of covered eyes were bigger than control eyes. Before and after cover, the diopter were 3.52±0.79 D and-2.88±1.61 D respectively, and there were statistically significant differences(P <0.05). Before and after cover, the eye axis were(7.32±0.18)mm and(8.60±0.20)mm respectively. The eye axis increased obviously and there were statistically significant differences(P <0.05). Compared with control group, the sclera organization morphology of dosing group improved obviously. The apoptosis rate of control group of control eyes and cover eyes were 13.43±3.50 and 85.71±12.75 respectively. The apoptosis rate of dosing group of cover eyes was 41.43±8.75. The difference had statistically significant(P <0.05). Tem observations show: Control group sparse form sleep deprived eye scleral collagen fiber distribution, collagen fiber diameter reduced obviously, gap between fiber increased. Sclera apoptosis populations, Control group, treatment group comparison eye scleral collagen fiber distribution uniformity, no significant changes in fiber diameter extracellular matrix. Did not see a typical apoptotic cell. Treatment group form sleep deprived eye scleral collagen fiber distribution is dense, collagen fibers decreased diameter is not obvious, fiber gap between mild increase. The apoptotic cells visible individual. Conclusions: Nourishing blood and Tonifying kidney decoction can markedly improve the sclera morphology of guinea pig form deprivation myopia, and significantly reduce the sclera cell apoptosis. Reducing sclera cell apoptosis may be the mechanism which Nourishing blood and Tonifying kidney decoction protecting the sclera cell.
