Objective:To explore the dynamic changes of activator protein 1(AP1)and colla gen I expression and analyze the relationship between them in myopic sclera coll agen remodeling.Methods:Selected 75 one-week-old guinea pigs,Randomly extracted 50 of them to establish the form deprivation myopia group(FDM group),the induction meth od was to cover the left eye of guinea pigs with head covering.The induction t ime was 2 weeks,4 weeks,4 weeks,1 week(4 /-1 weeks)and 6 weeks respe ctively.;and right eyes of them as the self-control group;eyes of the 25 untreated guinea pigs were referred to as the normal control group.Measured and recorded diopter and axial length of all guinea pigs at different time points.Western blot and immunohistochemistry were used to detect the dynamic changes of AP1 an d collagen I protein expression in the sclera of guinea pigs,Reverse transcriptionpolymerase chain reaction(RT-PCR)was used to detect the dynamic changes of AP1 and collagen I m RNA expression in the sclera of guinea pigs,the relationshi p between them was analyzed.Results:The diopter of the guinea pigs in the normal control group,the self-cont rol group and the FDM group were all hyperopic before covering eyes(all at P> 0.05).After 0,2,4,6 and 4/-1 weeks of form-deprivation,the diopter in the F DM group was gradually changed(from 2.09±0.31 D to-1.23±0.68 D,-4.17±0.59 D,-7.07±0.55 D and-2.67±0.59 D,respectively,p<0.05),while the axial length was gra dually increased(from5.93±0.38 mm to 6.62±0.37 mm,7.30±0.35 mm,7.99±0.31 mm and 6.97±0.32 mm,respectively,p<0.05).Compared with the normal control group a nd the self control group,the diopter of the guinea pigs was significantly changed in the FDM group after covering,and the axial length was also increased sign ificantly(all at P<0.05).With the prolongation of occlusion,,the protein expressio n of AP1 and collagen I in the FDM group were gradually weakened(all at p<0.05),the m RNA expression of AP1 and collagen I in the FDM group were also gradually down-regulated(all at p<0.05),and the correlation between them was p ositive,while the control groups had no obvious change in each index(all at p>0.05).Conclusions:These results suggest that AP1 can express in the sclera,and may be an important transcription factor involved in the regulation of collagen I synthesis and degradation during myopic scleral remodeling.Its specific mechanism needs further study.
