Objective:Study on hydrogen rich liquid on acute pancreatitis in rats serum endotoxin, TNFalpha, IL-6 level and the effect of pancreatic tissue, and to explore the mechanism of hydrogen rich liquid on acute pancreatitis and the therapeutic effect.Methods:weight in 250~300g, were randomly divided into 3 groups, respectively, the sham operation group(A group), normal saline group(group B), hydrogen rich water group(C group), 15 rats in each group, fasting treatment 24 h began to establish the model of acute pancreatitis in rats. Hydrogen rich water group and saline group rats pancreatic edema hyperemia, local dark red, and scattered bleeding points for modeling success, were given the same amount of saline and hydrogen rich water, sham operation group only slightly changes the pancreatic several times. After fasting rats after 24 H of treatment by intraperitoneal injection of urethane anesthesia. Into the abdomen through upper abdominal incision, to ensure no damage to vascular clipping hilar biliary and pancreatic duct and duodenum. Injection of 5% sodium taurocholate to push through the pancreatic duct, the dose of 0.1ML/1009, bolus injection speed is 0.2ML/MIN, after injection to maintain the injection pressure of 4MIN, waiting for.. After the success of modeling by gastric perfusion device according to the 125ML/KG/D, given saline or hydrogen rich water. Each group at first, 3, 7 day of 3 time points respectively, 5 rats were portal vein puncture whole blood 2ml, stay low temperature centrifugation, and then suck out the supernatant, ensure the standby keep in the-20 degrees in the refrigerator, resection of pancreatic tissue, and by cervical dislocation in material after sacrificed accordingly rat pancreatic tissue, making pathological sections in accordance with the conventional method, pathological changes of pancreatic tissue of rats were observed by HE staining under the microscope, and plasma endotoxin was measured by colorimetry with shark reagent azo, using enzyme linkedimmunosorbent assay(ELISA) for determination of serum TNF- alpha, IL-6 concentration,observe the general reaction of rats in each group during the whole experimental process.Results:(1) the reaction of A group rats is sensitive, fur smooth, gradually increasing body weight, respiration, in good condition; B rats after modeling, sluggish, unresponsive, limbs curled up, eyes dull; rats in group C after treatment, compared with the previous action reaction has improved, but the eyes are dull, dull.(2) each time point B, C group of endotoxin, TNF- alpha, IL-6 contents were significantly higher than those in A group(P < 0.05), the content of three components of C group were significantly lower than that of B group(P < 0.05), C group, TNF- alpha, IL-6of endotoxin in 12 h, 24 h, 48 h three time points were decreased while.(3) in pancreatic pathology in each time point in C group, B score was higher than that in A group(P < 0.05), in 24 h, 48 h, C group was significantly lower than that in B group the score(P < 0.05). The B group at three time points is increased, while the scores of C group called smooth.Conclusion:(1) in the pathogenesis of early acute pancreatitis, endotoxin in serum increased sharply, to promote a large number of proinflammatory cytokine synthesis and secretion of pancreatic tissue, eventually leading to severe damage;(2) in the process of acute pancreatitis, IL-6 in serum and TNF- levels will rise sharply, so IL-6 and TNF- levels can be used as a detection index of acute pancreatitis;(3) in the pathogenesis of acute pancreatitis, body endotoxin, IL-6 and TNF- levels can rise sharply, pancreatic tissue also suffered serious damage, hydrogen rich water can significantly lower serum endotoxin, IL-6 and TNF- levels, so as to realize the protective effect on pancreatic tissue.
