Objective:1.To explore the impact of electro-acupuncture on the sleep duration for(nembutal)sleep-deprived rats.2.To explore the impact of electro-acupuncture on changes of weight and behavior changes for PCPA sleep-deprived rats in open field test.3.To explore the impact of electro-acupuncture on the expression of mi RNA-132 in hypothalamus, as well as the expression of three key genes(Me CP2, P300 and JARID1A) and two biological clock genes(Per1 and Per2) for sleep-deprived rats.4.To summarize and probe into the functional mechanism for curing insomnia with electro-acupuncture.Methods:1. Observe changes in the rats’ sleep duration between control group and electro-acupuncture group by keeping a record of sleep-deprived rats’ sleep duration.2. Divide 32 Specific-pathogen-free Wistar rats randomly into four groups: control group, model group, diazepam group and electro-acupuncture group. The rats will be deprived of sleep by means of PCPA intraperitoneal injection two days in advance. The rats in diazepam group will receive intraperitoneal injection of diazepam for 5 consecutive days; while the rats in electro-acupuncture group will receive a 20-minute electro-acupuncture remedy on Baihui and Shenting for 5 consecutive days. After that, open field tests, designed to test crossing score,rearing score and pace time in center, will be held for those groups respectively. Hereupon Real-time PCR will be adopted to detect mi RNA-132 and changes in the expression of its target genes(Me CP2, P300, JARID1A) and circadian genes(Per1 and Per2).Results:1. The sleep duration of electro-acupuncture group lasts much longer than that of control group,which manifests significant statistical difference(P<0.05); 2. The rats in control group gain more weight than those in the other three groups; the rats in diazepam and electro-acupuncture group gain more weight than those in model group after receiving remedy, whereas the rats in electro-acupuncture group show more obvious increase in weight. All the differences manifest statistical significance(P<0.05).3. In the open field test, both the crossing score and the rearing score of the rats in electro-acupuncture group show downward trends compared with model group, while the pace time in center shows an upward trend. These three parameters all manifest statistical significance(P<0.05).4. After the rats being deprived of sleep, the expression of mi RNA-132 increases(P<0.01) and then it decreases to normal level after the rats receiving electro-acupuncture remedy. The difference manifests statistical significance(P<0.05); After the rats being deprived of sleep, the expressions of target genes(Me CP2, P300, JARID1A) and circadian gene(Per1) for mi RNA-132 decrease(P<0.05) and then they increase to normal level, even over-express, after the rats receiving electro-acupuncture remedy. The differences manifest statistical significance(P<0.05); After the rats being deprived of sleep, the expression of circadian gene(Per2) decreases(P<0.05) and then it shows an unremarkable increase after the rats receiving electro-acupuncture remedy. The difference is of no statistical significance(P>0.05).Conclusion:1. Electro-acupuncture remedy in Baihui and Shenting contributes to calming and sleep.2. PCPA intraperitoneal injection can make sleep-deprivation model of rats successfully.3. Sleep-deprivation will restrain the rats from gaining weight. While after the rats receiving electro-acupuncture remedy, their weight growth rate gradually returns to normal level, which indicates that electro-acupuncture remedy can adjust not only the rats’ sleep but also their digestive and endocrine systems.4. Sleep-deprivation will strengthen the excitability, aggressivity of the rats and weaken their cognitive competence. While electro-acupuncture remedy can do exactly the opposite to sleep-deprived rats. It is probable that electro-acupuncture remedy strengthens the cognitive competence of sleep-deprived rats through decreasing the overexpression of mi RNA-132.5. In sleep-deprived rats,the expression of mi RNA-132 increases and expression of Per1 reduces. Whereas electro-acupuncture remedy can reduce the overexpression of mi RNA-132 and improve the expression of circadian gene Per1. It is probable that the mechanism for improving the sleep state of sleep-deprived rats by means of electro-acupuncture remedy is related to the changes of mi RNA-132 and Per1 expression. |
