| Objective: To explore the mechanism of tumor inhibition and angiogenesis by investigating the effect of the Shenqi Antitumor Decoction on the liver cancer in mice.Methods:Select 60 Kunming mice which weight 20±2g. By using the method of axillar y injection of tumor strain, inoculated mice with H22 hepatoma ascites cells.After modelin g, the mice were randomly divided into six groups and 10 rats in each group.They modell gro up, positive control cisplatin group, low dosage Shenqi Anti-tumor Decoction group, high do sage Shenqi Anti-tumor Decoction group, low dosage Shenqi Anti-tumor Decoction combine d with cisplatin group and high dosage Shenqi Anti-tumor Decoction combined with cisplati n group.Second days after grouping, start treatment that mainly for the groups of Shenqi Ant i-tumor Decoction and the groups of Shenqi Anti-tumor Decoction combined with cisplatin. L ow dosage of the drug concentration was 0.5g crude drug /ml, and high dosage of the drug co ncentration was 1g crude drug /ml. Daily dosage for mice are 0.4ml/d.Cisplatin was injected i nto abdominal cavity and the dosage was 0.05mg/0.2ml/d. The continuous treatment of mic e was for 14 days.For other groups dosage: 0.05mg/0.2ml, two times a week. After t wo weeks, the blood was taken from the abdominal aorta, and the tumor tissue and s pleen were executed after the removal of the neck..During treatment observed and recor d(1)the condition of mice;(2) the spleen index,tumor weight and damps the lump rate of tum or;(3)the change of tumor specimen and tissue morphology;(4) the concentration of IL-4,IL-2,IFN-γ in serum;(5)the gene expression of the angiogenesis factor VEGF,b FGF,TGF-β,M MP13;(6) the expression of MMP13,VEGF,b FGF,TGF-β protein.Results:(1)Compared with the model group, the differences of weight in each treatment group were obvious, the difference was statistically significant(P<0.05).The animal’s general condition in Shenqi Anti-tumor Decoction combined with cisplatin group is better than that in the positive group.(2)For the inhibitory effect of H22 liver cancer, in high dosage traditional Chinese medicine it reached 35% and in low dosage of traditional Chinese medicine it was21%, and in positive control group it was 39%. Compared with pure Chinese traditional medicine group and cisplatin positive group, tumor inhibition effect in high dosage traditional Chinese medicine combined with cisplatin group was obvious.the difference was statisticallysignificant(P<0.05).(3)For the spleen index, comparing with the model group, it increased in the other groups. the difference had statistically significant(P<0.05);Compared with that in positive group. the spleen index in Shenqi anti-tumor decoction combined with cisplatin group and high dosage pure Chinese medicine group increased significantly. The differences had statistical significance.(P<0.05)(4) Compared with the model group, the concentration of IL-4 in serum of mice in the other groups all reduced, and the concentration of IL-2 and IFN-γall increased. The concentration of IL-2 and IFN-γ in serum of mice in pure Chinese medicine group was higher than that in cisplatin medicine group. The differences had statistical significance.(P<0.05)(5)Pathological results showed: in the traditional Chinese medicine groups the tumor cells vary in size and the number of tumor cell were more, and nuclear staining was not uniform. Tumor cell density decreased in Chinese medicine combined with cisplatin groups. Necrotic degeneration increased and there were many connective tissue overgrowth.(6)Compared with the model group, the protein and m RNA expression of VEGF, b FGF and TGF-β MMP13 in the positive control group, high dosage of Chinese medicine group and traditional Chinese medicine combined with cisplatin group was reduced.Compared with the positive control group, the expression m RNA of traditional Chinese medicine combined with cisplatin group was low, the difference was statistically significant(P<0.05).Conclusion:Shenqi Anti-tumor Decoction can inhibit the proliferation of H22 hepatocell ular carcinoma cells. And it has synergistic effect with cisplatin. The mechanism can b e summarized as the Shenqi Anti-tumor Decoction can increase the body’s immunity. A s well as the influence on tumor new angiogenic factors and generation of tumor angi ogenesis is its important role. |
