Objective: To research the expressions of cyclooxygenase-2(COX-2)、 X-ray repair cross complementing group 1(XRCC1) 、 ras association domain family 1(RASSF1) and their ralationship with clinicopathological characteristics and radiosensitivity and efficiency of esophageal squamous cell carcinoma(ESCC).Methods: The expression of the expression of COX-2 、XRCC1 and RASSF1 in 76 patients with ESCC who underwent radiotherapy alone were detected by immunohistochemistry method. Their relationship with clinicopathological characteristics and efficiency of radiotherapy for ESCC was analyzed by X2 test, Spearman Correlation analysis, Kaplan-Meier survival analysis, Log-rank test and Cox regression analysis.Results:1. It was found that the positive rate of COX-2、XRCC1 and RASSF1 in 76 ESCC tissue were 81.6%,52.6% and 59.2%, respectively. Spearman rank correlation analysis showed that between two expression in esophageal squamous carcinoma with no correlation(P > 0.038). 2. COX-2 protein expression of patients with esophageal squamous cell carcinomas on T stage, N stage and the clinical stages, the difference was statistically significant(P < 0.05); but in ESCC of the sex, age, tumor location, differentiation degree and the lesion length, there was no statistically significant difference(P > 0.05). XRCC1 protein expression in patients with ESCC of the sex, age, tumor location, T stage, N stage, clinical stage, differentiation degree and the lesion length, there was no statistically significant difference(P > 0.05). RASSF1 protein expression of patients with ESCC on clinical stages, the difference was statistically significant(P < 0.05); and on sex, age, tumor location, T stage, N stage, differentiation degree and the lesion length,there was no statistically significant difference(P > 0.05). 3. The relationship between COX-2, XRCC1 and RASSF1 protein expression and radiosensitivity and prognosis The RR of COX-2 protein expression negative group was 92.8%(13/14), higher than 66.1%(41/62) positive expression group, there was statistically significant difference(X2 = 3.967, P < 0.05); Cox-2 protein positive expression and esophageal cancer radiotherapy recent curative effect is negative correlation relationship(r = 0.228, P = 0.047). The RR of XRCC1 protein expression of negative group was 77.8%(28/36), the positive expression of group 65%(26/40) slightly increases, but there was no statistically significant difference(X2 = 1.504, P > 1.504). The RR of RASSF1 protein expression positive group was 80%(36/45), higher than 58.1%(18/31) negative expression group, there was statistically significant difference(X2 = 4.294, P < 0.05); RASSF1 protein positive expression and esophageal cancer radiotherapy recent curative effect is positively related to relationship(r = 0.238, P = 0.039). The expression of COX-2 was correlated with radiosensitivity of ESCC, but is not independent factors. Full set of complete remission(CR) in 41 cases(53.9%) and partial response(PR) in 13(17.1%), SD for 18 cases(23.7%), and PD for 4 cases(5.1%). The overall 1, 2, 3, 4 years of survival rates for ESCC after radiotherapy were 62%, 37%, 18%, 13%, and the median surial was 17 months. Univariate analysis showed that T staging, overall response rate, RASSF1 positive expression and RASSF1 positive + XRCC1 negative(P= 0.034, 0.000, 0.004, 0.008)associated with prognosis. While gender, age, tumor location, N stage, clinical stage, differentiation degree, the length of lesion, COX-2 expression, XRCC1 expression, expression of COX-2 and XRCC1 and negative, negative + RASSF1 express positive COX-2 expression, as well as RASSF1 positive + COX-2 negative + XRCC1 negative, has no association with radiation prognosis(P > 0.05). Multivariate analysis showed that overall response rate and RASSF1 positive expression radiation as independent prognostic factors(P =0.000 and 0.026), and its associated OR values were 3.724 and 2.065。.Conclusion: The expression of COX-2 was correlated with radiosensitivity of ESCC, but is not independent factors.XRCC1 protein negative expression of overall response rate higher efficient than positive expression, to some extent hints XRCC1 protein negative expression of patients may benefit from radiation therapy.Positive expression of RASSF1 associated with esophageal radiation sensitivity, is an independent prognostic factor.The alleviate after radical radiotherapy, RASSF1 positive expression has good prognosis of radiotherapy.
