Objectives 1 Discuss different obese pregnant women during pregnancy on an empty stomach in the peripheral blood plasma Ghrelin and Obestatin levels change, and the correlation of the lipid metabolism and sugar metabolic abnormalities. 2 Obese pregnant women Obestatin levels in umbilical cord blood and neonatal the relationship between body mass index. 3 Explore Ghrelin and Obestatin protein expression in placenta. Methods 1 In diagnostic criteria, inclusion criteria and exclusion criteria, obese pregnant women and the fat of pregnant women, the 50 cases, respectively, take different maternal peripheral blood on an empty stomach, obese pregnant women during pregnancy childbirth in umbilical cord blood, and measuring neonatal body mass index, t two groups of pregnant women in childbirth are to return part of the placenta tissue. 2Ghrelin, Obestatin levels was evaluated by radioimmunity method. 3 Maternal fasting glucose was evaluated by glucose oxidase method. 4 Use enzyme-linked immune sorbent assay determination of maternal fasting insulin. 5 Maternal serum total cholesterol was evaluated by cholesterol oxidase method. 6 Use ultraviolet enzymatic determination of maternal serum triglycerides. 7 Use direct determination method of determination of high-density lipoprotein cholesterol. 8 Use immune histochemical methods about Ghrelin,Obestatin protein expression in the tissue of the placenta. 9 For the statistical analysis using SPSS 13.0 statistical software package, Value to sx ±, two groups of data comparison using the t test, variables in the group using Pearson correlation analysis. Results 1 Obese pregnant women, in the early stages of pregnancy at mid and late pregnancy peripheral blood plasma Ghrelin levels respectively 230.32±10.46 pg/ml 、250.18±14.17 pg/ml 、 231.94±11.59 pg/ml. Mother in Ghrelin levels in the blood:Normal pregnant women, in the early stages of pregnancy pregnant metaphase and late pregnancy peripheral blood plasma Ghrelin levels respectively 317.56±15.22 pg/ml 、349.64±12.25 pg/ml、325.02±12.75 pg/ml. The results showed, two groups of pregnant women on an empty stomach in the peripheral blood plasma Ghrelin levels are a downward trend for the early stages of pregnancy, mid pregnancy began to rise up to peak, late pregnancy and fell, but higher than the early stages of pregnancy difference was statistically significant(P<0.05). But with the change trend of plasma Ghrelin levels in obese pregnant women during pregnancy than non-obese women significantly, the difference was statistically significant(P<0.05). 2 Obese pregnant women in the early, middle and late pregnancy peripheral blood plasma Obestatin levels respectively284.52±12.39 pg/ml 、 260.83±12.89 pg/ml 、 229.81±40.82 pg/ml. Mother Obestatin levels changes in the blood: Normal group of pregnant women in the early, middle and late pregnancy peripheral blood plasma Obestatin levels respectively 370.31±14.88pg/ml、324.73±15.04 pg/ml、263.37±44.70 pg/ml. The results showed, two groups of pregnant women on an empty stomach Obestatin levels in peripheral blood plasma with gestational age development trends to decrease, the difference was statistically significant(P<0.05). But with obese pregnant women during pregnancy in the peripheral blood plasma Obestatin levels are lower than the obese pregnant women, and obese pregnant women the whole Obestatin levels decrease trend more obvious, the difference was statistically significant(P<0.05). 3 Obese pregnant women on an empty stomach Obestatin levels in peripheral blood plasma and negatively correlated to the level of FINS,HOMA-IR the difference was statistically significant(r=-0.86,P<0.05 and r=-0.82,P<0.05). 4 Obese pregnant women late in pregnancy, fasting plasma Obestatin levels and negatively correlated with TG, the difference was statistically significant(r =-0.88,P<0.05 and r=-0.74,P<0.05); Late pregnancy and negatively correlated with LDL, the difference was statistically significant(r=-0.75,P<0.05). 5 In the normal group and obesity group of neonatal umbilical cord blood Obestatin levels respectively215.34±15.26 pg/ml 、 102.58±16.32 pg/ml, compared with normal group, obese Obestatin levels in the neonatal umbilical cord blood, the difference was statistically significant(P<0.05). 6 Obese compared neonatal body mass index and non-obese group, birth weight, abdominal skinfold increase obesity group, the difference was statistically significant(P<0.05). 7 Obestatin levels in obesity group of neonatal umbilical cord blood and negatively correlated with birth weight, abdominal skinfold, the difference was sta tistically significant(r=-0.789, P<0.05 and r=-0.840, P<0.05). 8 Immunohistochemical found in the two groups of pregnant women placenta trophocyte Obestatin and Ghrelin protein expression. Conclusions 1 Two groups of pregnant women in different fasting plasma Ghrelin levels in peripheral blood during pregnancy has certain change, and obese pregnant women are not obese pregnant women change tendency is more significant. 2 Two groups of pregnant women on an empty stomach peripheral blood plasma Obestatin levels were declined with the increase of gestational age, and obese pregnant women is far greater than non obese pregnant women. 3 Obese pregnant women Ghrelin, Obestatin and lipid metabolism disorders, sugar metabolic abnormalities have a certain correlation.4 Obese pregnant women Ghrelin and Obestatin levels in umbilical cord blood and neonatal weight and index system have certain relevance. 5 Two groups are Ghrelin and Obestatin protein expression in placenta of pregnant women.
