| Objective Research the in vitro antimicrobial activity of schizandrin A on Enterococcus faecalis biofilm,and to investigate the antibacterial effect is completed through the effect on the expression of virulence factors of transcription or interference.Methods The establishment of the sample of Enterococcus faecalis biofilm model in vitro by holes in 96, the inhibition rate of the MTT was detected by the method of detecting the biofilm of the feces. Establish biofilm model in 6-well plates, laser confocal microscope(CLSM) was observed to observe the changes of the different concentrations of drug in the biofilm after the biofilm. Select theappropriate concentration of drug-, the use of RT-PCR technique to detect the expression of Enterococcus faecalis virulence factor.Results Schisandrin and Enterococcus faecalis biofilm inhibitory effect was positively related to the trend, that is with the increase of drug concentration, its inhibitory effect increaseed. CLSM testing results showed that with the increase of drug concentration, the establishment of viable bacteria biofilm model in proportion to reduce, dead bacteria signifycantly increased the proportion of. The results of RT-PCR showed that the expression of virulencefactors srtCã€espã€ebpR decreased with the increase of drug concentration gradient, at the concentration of 500 ug/ml and higher, the level of mRNA expression inhibited.Conclusions Schizandrin A in vitro to inhibitory effect on Enterococcus faecalis,and express the process of this experiment selected virulence factors srtCã€espã€ebpR inhibited. |
