Objectives:The experimental research is aimed to explore the effects of Xiaoaijiedu decoction on antitumor by intervening T regulatory cell in the microenvironment of hepatocellular carcinoma.Methods:In SD rats model,rat hepatocarcinoma model is induced by DEN aqueous solution except the normal group.Physiological saline was administered to the control group,Xiaoaijiedu decoxtion was administered to the drug groups as the prescribed does.Observing the expression of Foxp3 in liver,spleen and thymus.Detecting Foxp3 in liver by immunohistochemical techniques; Tseting T regulatory cells in spleen and thymus using flow cell technology; Detecting Foxp3 protein in spleen tissue homogenates by Wserern-blot technology.Results:(1)Compared with the control group,6.25,12.5 and 25g/kg of Xiaoaijiedu decoction can reduce tumor weight,tumor suppressor rate is 49.13%,60.20% and 70.93%. (2) Compared with the control group, the groups of Xiaoaijiedu decoction reduce Foxp3 in liver and the greater the does,the Foxp3 expresses less. (3) Compared with the normal group, the control group can considerably increase the expression of CD4 and Foxp3 in spleen (P<0.05);Compared with the control group,6.25,12.5 and 25g/kg of Xiaoaijiedu decoction reduce the expression of CD4 and Foxp3 in spleen(P<0.05); Compared with the normal group, the control group and 6.25 of Xiaoaijiedu decoction increase the expression of CD4 and Foxp3 in thymus(P<0.05); Compared with the control group,6.25.12.5 and 25g/kg of Xiaoaijiedu decoction reduce the expression of CD4 and Foxp3 in thymus(P<0.05) (4)Compared with the normal group,the control group increases the relative gray value of Foxp3 (P<0.05);Compared with the control group,6.25g/kg of Xiaoaijiedu decoction reduces the relative gray value of Foxp3 (P>0.05); Compared with the control group,12.5 and 25g/kg of Xiaoaijiedu decoction reduce the relative gray value of Foxp3 (P<0.05).Conclusion:(1)Xiaoaijiedu decoxtion can reduce the tumor weight and has a good antitumor effect. (2)Xiaoaijiedu decoction can reduce the expression of Foxp3 in liver, thymus and spleen. Xiaoaijiedu decoction inhibites the growth and function of regulatory T cell to suppress the occurrence and development of tumor. |
