Objective:Part 1:To study the expression of B7-H4 in gastric cancer and analyse the relationship between B7-H4 and the clinical pathological parameters of patients with gastric cancer.Part 2:To study the expression of Foxp3 in gastric cancer and analyse the relationship between Foxp3 positive regulatory T cells and the clinical pathological parameters of patients with gastric cancer.To statistical analyse the correlation between B7-H4 and Foxp3 positive regulatory T cells,analyse the affect of B7-H4 and Foxp3 positive regulatory T cells to the tumor associated macrophages,and analyse the effect of B7-H4 and Foxp3 positive regulatory T cells to the prognosis of patients with gastric cancer.Methods:Part 1: The expression of B7-H4 was detected by immunohistochemistry(SP method)in 120 cases of gastric adenocarcinoma tissues.Then we analyzed the relationship between the expression of B7-H4 and clinical pathological parameters.Part 2: The expression of Foxp3 positive regulatory T cells were detected by immunohistochemistry(SP method)in 120 cases of gastric adenocarcinoma tissues.The relationship between the expression of Foxp3 positive regulatory T cells and clinical pathological parameters was analyzed.Then we investigated the effect of B7-H4 expression and Foxp3 positive regulatory T cells infiltration to the prognosis of patients in gastric cancer tissues,and impact to the tumor associated macrophages Results:Part 1: The immunohistochemistry results showed that B7-H4 was found in 80 of 120 cases of patients with gastric adenocarcinoma(66.7% positive rate).The expression of B7-H4 was positively correlated to lymph node metastasis and pathological staging(P<0.05),but had no correlation with patient’s sex,age,histological type and the invasion depth(P>0.05).Part 2: Foxp3 were expression in the nuclear of regulatory T cells,and the positive rates were 65%.Foxp3 expression were positively correlated to lymph node metastasis,pathological staging and invasion depth(P<0.05),but had no correlation with patient’s sex,age and histological type(P>0.05).The expression of B7-H4 and Foxp3 positive regulatory T cells were positive correlation in gastric adenocarcinoma(r=0.675,P=0.001).Compared with low(or negative)B7-H4 expression or low Foxp3 positive regulatory T cells count,the prognosis of patients in high B7-H4 expression and increased Foxp3 positive regulatory T cells count was worse.B7-H4 and Foxp3 positive regulatory T cells affected tumor associated macrophages.Conclusion:The upregulation of B7-H4 and Foxp3 positive regulatory T cells in gastric cancer may be correlated with gastric cancer metastasis and prognosis.The expressions of B7-H4 and Foxp3 positive regulatory T cells were positive correlation in gastric cancer,both of their high expression can upregulate tumor associated macrophages.The expressions of B7-H4,Foxp3 positive regulatory T cells and tumor associated macrophages affect each other,and they influence on the prognosis of patients. |
