Objective:(1)The submerged fermentation medium of Agaricus blazei Murill was optimized to increase mycelial biomass and extracellular polysaccharide production.(2)To examine the protective effect of ABM cultured by alcohol induced liver injury in mice model.(3)Protecting liver dosage form of Agaricus blazei Murill was researched through studying the preparation technology and quality standard.Methods:(1)The medium composition of Agaricus blazei Murill was optimized using response surface methodology with Box-Behnken on mycelial biomass and extracellular polysaccharide(EPS) production, which impact factors were concentrations of potato, bran and glucose.(2)The model of mice with chronic alcoholic hepatic injury was intragastrically perfused with high concentration alcohol to measured the content of ALT and AST in serum and hepatic tissue.(3)Formulation composition and preparation technology of protecting liver capsule of Agaricus blazei Murill were determined by investigating the hygroscopicity and liquidity of excipients.(4)Quality standard of protecting liver capsule of Agaricus blazei Murill was confirmed through determining the extracellular polysaccharide production in capsules.Results:(1)The model predicts to gain a maximal mycelial biomass and extracellular polysaccharide at 1.047 g/100 m L,and 0.367 g/100 m L,respectively,when the potato is 29.88 g/100 m L,the glucose is 1.01 g/100 m L,and the bran is 1.02 g/100 m L.(2)The levels of ALT and AST were decreased respectively 18%~32%,15%~23% in sample groups compared to model group. There were no or a small amount of dead cells and inflammatory lesions by examining liver pathologic histology in mice.(3)Microcrystalline cellulose was chosen as absorbent through investigating the hygroscopicity and liquidity of excipients. And the ratio of Microcrystalline cellulose was 1:1(W/V).(4)Extracellular polysaccharide production determined in capsules was 3.03%. The results of the checks of this preparation were in line with the related provisions of 2010 edition of “Chinese Pharmacopoeia”.Conclusions:(1)Response surface methodology is able to predict the mycelial biomass and extracellular polysaccharide(EPS) production of Agaricus blazei Murill Liquid fermentation.(2)Agaricus blazei Murill with liquid fermentation had certain protective effect on alcoholic liver injury in mice.(3)The preparation technology and quality standard of rotecting liver capsule of Agaricus blazei Murill were determined.
