Influences Of Banxia Xiexin Decoction And Split Prescriptions On Th1/Th2 Cytokines Of Rats With Ulcerative Colitis

Objective: To explore immunomodulatory mechanism of Banxia Xiexin Decoction for Ulcerative Colitis(UC) rats model, as well as the regulation function of sweet-tonifying, bitter-cooling and pungent-warming drugs in the prescription for Th1/Th2.Methods:(1)We collected oral herbal medicine formula based on clinical effects on UC and analyzed effective therapies and prescriptions. And the Banxia Xiexin Decoction was selected for split study according to prescription characteristics with Traditional Chinese Medicine Inheritance Support System(TCMISS).(2)We established rats model of UC with TNBS/ethanol mixing.(3) We observed anti-inflammatory functions of Banxia Xiexin Decoction and split prescriptions on UC according to general conditions, CMDI scores and pathologic changes.(4) Elisa was applied for contents test of IFN-γ, IL-4, IL-6 and IL-10 in serum.(5) Immunohistochemistry(IHC) was adopted for testing JAK2 and STAK3 protein expression in colon tissues.Results:(1) Prescriptions data analysis of treating UC based on clinical effects, Qi-tonifying, Heat-clearing, Astriction, Warming-interior drugs were used more frequently and the high-frequency drugs and core combination showed therapies of Strengthening Spleen and Tonifying Qi, Clearing Heat and Drying Dampness as well as Warming the Interior and Dispelling Coldness were common for UC. The core drugs corresponded to Banxia Xiexin Decoction prescription chracteristics.(2)Compared to the control group, the model group got higher CMDI and TDI scores(p<0.05); compared to the model group, the Banxia Xiexin Decoction, cooling-tonifying group and sweet-tonifying group got lower scores(p<0.05).(3) For IFN-γ in serum, compared to the control group, the model group had increased contents with statistical significance(p<0.05); compared to the model group, the warming group, tonifying group, cooling-tonifying group and Banxia Xiexin Decoction group had decreased contents with statistical significance(p<0.05). For IL-4, compared to the control group, the model group had decreased contents with statistical significance(p<0.05); and compared to the model group, the compared to the control group, the model group had decreased contents with statistical significance(p<0.05);, cooling-tonifying group, tonifying group and cooling group had increased contents with statistical significance(p<0.05). For IL-6, compared to the control group, the model group had decreased contents with statistical significance(p<0.05); the cooling-warming group, warming-tonifying group, cooling-tonifying group and Banxia Xiexin Decoction group had decreased contents(p<0.05). For IL-10, compared to the control group, the model group had decreased contents(p<0.05); and compared to the model group, Banxia Xiexin Decoction group, cooling-tonifying group, warming-tonifying group and tonifying group had increased contents(p<0.05). For ratio of IFN-γ/IL-4 and IL-6/IL-10, compared to the control group, the ratio of the model group increased(p<0.05); compared to the model group, the ratio of the Banxia Xiexin Decoction group, sweet-tonifying group and cooling-tonifying group decreased(p<0.05). And for JAK2 and STAK3 protein expression in colon mucosa, compared to the control group, the model group had increased expression contents with statistical significance(p<0.05); and compared to the model group, Banxia Xiexin Decoction group, the cooling group and cooling group and cooling-warming group had decreased contents but with no statistical significance.Conclusion:(1) Banxia Xiexin Decoction and split prescriptions show favorable anti-inflammatory effects on UC, among which the whole prescription has the most significant effects.(2)Banxia Xiexin Decoction comprehensively regulates rats’ immune microcircumstances through reducing Th1 and raising Th2 immunocytokines, and then promotes inflammation healing prognosis.(3)The sweet-tonifying drugs regulate immune balance through reducing IFN-γ and raising IL-4 and IL-10. And the sweet-tonifying combined with the pungent-warming drugs regulates Th1 through reducing IL-6; the sweet-tonifying combined with the bitter-cooling drugs regulates Th1/Th2 immune balance through reducing IL-6 and raising IL-4 and IL-10. While the combination of the sweet-tonifying, pungent-warming and bitter-cooling drugs shows the optimum effects on regulating Th1/Th2 balance.(4)The immunocytokines regulating mechanism of each group doesn’t show any significant dependence on JAK/STAT access.
