Purpose:prepare Isolated and Cannulated Venules and TNF-alpha intervention venule albumin leakage models by using Microsurgery operation technique. By giving the endocytosis inhibitor and hydroxyethyl starch intervention, observe and compare the apparent permeability coefficient to study whether Hydroxyethyl Starch can make effect on venule albumin leakage, and the possible pathway it intervented.Method:step one:12 adult male rabbits (weight 2.5kg ± 0.5kg) randomly divided into saline group (control group,6) and hydroxyethyl starch group (experimental group,6) using random number table.Step two:the two groups underwent microsurgical operation preparation of Isolated and Cannulated Venules, with Fluorescein isothiocyanate labeled bovine serum albumin (FITC-BSA) as the tracer albumin, Determinate the apparent permeability coefficient as foundation value before the foundation of venule albumin leakage model, then use TNF-alpha to prepare venule albumin leakage models:1、Determinate apparent permeability coefficient of the control group while intervented by Saline, or saline+endocytosis inhibitor;2、Determinate apparent permeability coefficient of the experimental group while intervented by Hydroxyethyl Starch, or Hydroxyethyl Starch+endocytosis inhibitor;Step three:data analysis:1、compare apparent permeability coefficient of the two groups before and after infusion of TNF-alpha;2、compare apparent permeability coefficient between the groups with TNF-alpha intervention;3、compare apparent permeability coefficient of the two groups before and after using endocytosis inhibitor;4、compare apparent permeability coefficient between the groups with endocytosis inhibitor intervention;Results:1、The apparent permeability coefficients in two groups were increased after infusion of TNF-alpha, the difference was statistically significant (P< 0.05).2、after infusion of TNF-alpha, the control group’s apparent permeability coefficients are higher than the experimental group’s, the difference was statistically significant (P< 0.05).3、there are no significant differences (P> 0.05) of apparent permeability coefficients in two groups before and after using endocytosis inhibitors.4、after using endocytosis inhibitors, the control group’s apparent permeability coefficients are higher than the experimental group’s, the difference was statistically significant (P< 0.05).Conclusion:1、the production of venule albumin leakage model is successful.2、TNF-alpha may mainly increase the paracellular pathway causing venule albumin leakage.3、hydroxyethyl starch can inhibit the leakage of albumin on TNF-alpha induced venules.4、hydroxyethyl starch may mainly intervene the paracellular pathway.
