Objective:To elucidate the association of the familial aggregation of hepatocellular carcinoma(HCC) with HLA-DRB1*15allele as well as the expression level of Th1/Th2cytokines in Guangxi.Methods:With the same sexuality, age±5year, same ethnic group, same living environment, same habits and same HBV carried as paried condition,90members whose families have had two or more HCC patients (high-occurrence families) were selected as the case group, and90members whose families have no any cancer patient were selected as the controls. All the members without HCV infection. PCR-SSP was used to detect HLA-DRB1*15allele and ELISA was used to detect IFN-γ, IL-2, IL-4and IL-10.Results:(1)The frequency of the HLA-DRB1*15allele in the case group was higher than that in the controls (P<0.05);(2) The expression levels of IFN-γ-, IL-2in the case group was less than that in the controls (P<0.05), but the expression levels of IL-4, IL-10in the case group was less than that in the controls (P<0.05);(3) In the case group, the expression level of IL-2in the positive group of the HLA-DRB1*15allele was less than that in the negative group of the HLA-DRB1*15allele(P<0.05);(4) In the case group, the expression level of IL-10in the positive group of the HLA-DRB1*15allele was highter than that in the negative group of the HLA-DRB1*15allele(P<0.05);(5) To eliminate the influence of HBV infected, in the the case group, the positive of HBsAg in the negative group of the HLA-DRB1*15is the same with the positive of HBsAg in the positive group of the HLA-DRB1*15(P>0.05)Conclusions:HLA-DRB1*15allele and the imbalance of Thl/Th2cytokines might be associated with familial clustering of hepatocellular carcinoma in Guangxi, China. The imbalance of IL-2, IL-10might be due to the carrying of HLA-DRB1*15. Through the same approaches, these might lead to the phenomenon of familial aggregation of HCC in Guangxi.
