Objective:To analyse a possible link between MMP-2and the invasive growth of E. m tissue of liver in mice.Methods:The245healthy female mice of14-week-old were randomly divided into seven groups,35per group.Group A: blank control group; group B: negative control group; group C: model control group; group D:empty vector adenovirus(Mock) injection group; group E: scrambled MMP-2sequence adenovirus(Ad-SV)injection group; group F: siRNA against MMP-2adenovirus pretreatment injection group; group G: siRNAagainst MMP-2adenovirus(Ad-MMP-2) injection group. A, B and C group of mice were not intervene,while D, E, G and F groups were intravenously injected with empty vector adenovirus, Ad-SV, Ad-MMP-2and PBS. After100days of modeling, every group of mice were sacrificed to observe the growth of thehepatic E. m tissue, to calculated infection, cyst weight and reduced rates of hepatic E.m tissue in differentexprimental groups, to observe the changes of histomorphology with HE stain, to detect the apoptosis indexof the distant hepatocyte, near the puncture site or near the E. m hepatocyte and the inflammatory cellsurrounding the hepatic E. m tissue with TUNEL method, to detect the expression of MMP-2and TIMP-2in the serum of different groups mice with ELISA method, to determine the Caspase-3content of thehepatic E. m tissue, the near E. m liver tissue and the distant liver tissue in different groups mice withCaspase-3kit,and to determine the expression of Collagen â…£ in the pericystic layer of hydatid cysts indifferent experimental groups with immunohistochemistry method.Result:The livers of mice in groups A and B had no E. m tissue growth. Infection of hepatic E.m tissue ingroup C, D, E, F and G were72.4%,70.0%,71.4%,72.0%and70.8%, respectively, had no significantdifference among them (P>0.05). The quality of hepatic E. m tissue in mice had no significant differencecompared between the groups of C, D, E and F (P>0.05), but it showed a significant difference comparedwith group G, respectively (P<0.05). Reduced rates of hepatic E. m tissue had no significant differencecompared between the groups of D, E and F, but but it showed a significant difference compared withgroup G, respectively (P<0.05). The apoptotic indexes(AI) of distant hepatocyte were no significantdifference in all groups mice(P>0.05); the AI of hepatocyte near the E. m tissue compared to distanthepatocyte among the all inoculation hepatic E. m groups showed a significant difference (P<0.05); the AIof hepatocyte near the E. m tissue of group G was higher than group B while lower than the groups of C, D,E, F and they all showed a significant difference (P<0.05); the AI of the inflammatory cell surrounding thehepatic E. m tissue among the groups of C, D, E, F had no difference (P>0.05), while comparing the groupG with them respectively showed a significant difference (P<0.05). The expression of MMP-2had nosignificant difference compared between the groups of C, D and E (P<0.05), groups of C, D, E showedsignificant difference compared with A, B, F, G, respectively (P<0.05), group F showed significantdifference compared with C, D, E, G, respectively (P<0.05), group G showed significant differencecompared with C, D, E, F, respectively (P<0.05). The expression of TIMP-2had no significant differencecompared between the groups A, B, C, D, E, F, G (P<0.05). The rate of MMP-2/TIMP-2in group Gshowed significant difference compared between groups A, B, C, D, E, F, respectively (P<0.05). TheCaspase-3contents of the distant hepatic tissue in different group mice showed no difference (P>0.05), theCaspase-3content of the hepatic tissue near the puncture site in the group B showed a significant differencecompared with the groups of C, D, E, F, G, respectively (P<0.05), the Caspase-3content of the hepatictissue near the E. m in group C, D, E showed significant difference compared with its Caspase-3content ofthe E. m tissue, respectively (P<0.05), the Caspase-3content of the hepatic tissue near the E. m in groups C,D, E showed no significant difference(P>0.05), the Caspase-3content of the hepatic tissue near the E. m ingroups F, G showed significant difference compared with groups C, D, E, respectively (P<0.05), theCaspase-3content of the E. m tissue in group G showed significant difference compared with groups C, D,E, F, respectively (P<0.05). Collagen â…£ in the pericystic layer of hydatid cysts in group G showed significant difference compared with G\group C, D, E, F, respectively (P<0.05).Conlusion:MMP-2was highly expressed in the model of hepatic echinococcus multilocularis disease,Echinococcus multiloculars promote the tissue that surrounding it secreted MMP-2during the growthprocess in the host, MMP-2siRNA inhibited the MMP-2express in the host, which inhibited the invasivegrowth of E. m, therefore, MMP-2play an important role in the invasive growth of E. m in liver of host.1. When the expression of MMP-2was inhibited, the matrix degradation surrounding hepatic E. m wasinhibited, while hindering the invasive growth of hepatic echinococcus multilocularis.2. Reducing the rate of MMP-2/TIMP-2, which can inhibit the invasive growth of hepatic echinococcusmultilocularis.3. Inhibiting the expression of MMP-2in host, the apoptosis of liver and inflammatory cells surroundingthe hepatic echinococcus multilocularis were inhibited, which may play an important role in inhibition ofhepatic echinococcus multilocularis invasive growth.4. MMP-2may be a new target for treatment of hepatic echinococcus multilocularis disease.
