Objective:To study the2,4-D butylate on reproductive toxicity and its mechanism of malemice by observing the effects of2,4-D butylate on spermatogenesis in male miceã€Pathological changes of testis tissue and detected the Biological effect index whichassociated with toxicity in testis,to further reveal the potential reproductive toxicity2,4-D butylate arising from environmental pollution and provide scientific basis formale reproductive health.Methods:Forty-eight ICR male mice were randomly divided into four groups by weight,One was as a control group,the other three was as experimental groups:10mg/kg(1/40LD50)ã€20mg/kg(1/20LD50)ã€40mg/kg(1/10LD50),and twelve in each group.The2,4-D butylate was dissolved by corn oil,Intragastric administration was once aday for five weeks.After24hours of exposure, the mice were killed by Jin vertebral dislocation afterthe eyeballs were extracted and blood, Sperm count,and the marker enzyme activityof testis tissueã€the activity of antioxidant enzymes and ATPase and the content oflipid peroxidation product,the level of testosterone in serum were detected,to observethe pathological changes of testis of mice.The experimental data were analyzed by SPSS13.0and the results wereexpressed asx s, one-wayANOVA was used for among groups,and SNK test forcomparison between teo groups, the level of test: a=0.05,P<0.05was consideredstatistically significant.Results:1. Effects of2,4-D butylate on testicular and epididymal organ coefficients inmale miceThe organ coefficient of testis was gradually decreased as the dose increased,there was significant difference between the high dose group and the middle dose group with control (P<0.05); The organ coefficient of epididymal has a significantdifference between the high dose group with middle and the control group.2.Effects of2,4-D butylate on Sperm counts in male miceThe Sperm counts of epididymal was gradually decreased as the dose of2,4-Dbutylate increased, and the Sperm counts of epididymal in the middle and high dosegroup were significantly lower compared with the low dose group and controlgroup(P<0.05);The rate of sperm Aberration was gradually increased as the doseincreased,the difference between the high dose group with middle and control groupwith low dose group has statistical significance(P<0.05).3. The oxidative damage of2,4-D butylate on the testis in male miceThe activities of T-AOC was gradually decreased as the dose increased,thedifference between high dose group and other groups have statisticalsignificance(P<0.05); The activities of SOD was gradually decreased as the doseincreased,the difference between the each experimental group has statisticalsignificance (P<0.05);The content of MDA was gradually increased as the doseincreased, the difference between the high dose group and the control group hasstatistical significance (P<0.05).4. Effects of2,4-D butylate on the marker enzyme activity of testis tissue inmale miceThe activities of SDH in testis was gradually decreased as the dose of2,4-Dbutylate increased, there was significant difference between the high dose group withmiddle and the control group(P<0.05),and the difference between high dose groupand middle with low dose group has statistical significance(P<0.05);The activities ofLDH in the middle and high dose group were significantly lower compared with thelow dose group and control group (P<0.05),and the difference between high dosegroup and middle dose group has statistical significance(P<0.05).5. Effects of2,4-D butylate on activities of ATPase in the testis tissue of miceThe difference of the activities of Ca﹢﹢Mg﹢﹢-ATPase between the dose groupand the control group has statistical significance(P<0.05);The activities of Naï¹¢Kï¹¢-ATPase in the middle and high dose group were significantly lower than that of thecontrol and low dose group,and the difference had statistical significance(P<0.05).6. Effects of2,4-D butylate on testosterone level in serum of male miceThe level of T in serum was gradually increased as the dose increased, and the level of serum T in the middle and high dose group were significantly lowercompared with the low dose group and control group(P<0.05),and the differencebetween high dose group and middle dose group has statistical significance (P<0.05)7. Effects of2,4-D butylate on Pathologic change of testis in male miceThe pathological section of mouse testicular shown that: the spermatogenicepithelium of low dose group had no obvious change, but there are nuclearenrichment phenomenon in individual primary spermatocyte or secondaryspermatocytes; spermatogenic epithelium thinning, spermatogenic cells arranged indisorder, unclear levels, part missing, the lumen visible out in the seminiferousepithelium of spermatogenic cells, located near the lumen of mature sperm rare inmiddle and high dose group.Conclusions:This study shows that exposed2,4-D butylate have effects on Sperm counts inmale mice; with the oxidative damage of testis tissue;inhibition the activity of markerenzyme and serum testosterone of testis;reducing the level of testosterone inserum;effect the morphological structure of testis of mice. |
