objective: Rectal cancer is a common malignant tumor, operation is the main treatment method. But because of the special position ofrectal, the reccurence rate is high. The currently accepted The clinical stageand the distance between tumor and the anal are all major factors in judgingthe prognosis, but the same stage of the patients received the same treatment, the result is not the same, whether there are other factors that affect outcome? This is thought-provoking. In recent years, although the treatment of adjuvantchemoradiotherapy is introduced into this disease, survival rate of patientswere improved, but the mutility rate is still high. Y-box binding protein isubiquitousely expressed transcription factor, with a variety cellular functions. Many studies found it closed related with a wide variety of human malignanttumor, including developing of tumor, drug resistence, prognosis. However, there is little research on the relationship between YB-1and rectal cancer, especially the relationship with prognosis of rectal cancer, there had no reportabout it. So, our study want to examine the expression of Y-box bindingprotein1in tissues of rectal cancer, rectal cubular adenoma, rectitis, andanalysis the relevance to rectal cancer. And the same time, to collect theclinical information of patients and follow up them. Then, to investigate thedeveloping and prognostic role of YB-1in rectal cancer. Methods:1.Collected the clinical data of rectal cancer patients between2008and2009from the centry hospital of Mianyang, collected80cases of them. Then collected30rectal tubular adenoma cases and30rectitis cases.2.Collected80cases of rectal cancer patients with clinical information and follow-up data.3.Selected72rectal cancer’s tissues and30tissues of rectal tubular adenoma, rectitis respectively, to detect YB-1expression with immunohistochemicalmethod. All of them were verified by pathology4. Analysis the expressiondifference of YB-1among these three rectal tissues; and the relationshipbetween the level of YB-1and prognosis index (DFS or OS) wereanalysed.Results:1.The mean YB-1histological score(HSCORE) values forrectal cancer, rectal tubular adenoma and rectitis tissues were205.5,164.3and137.7respectively.2.There were72rectal cancer patients finished thefollow-up, the higher expression of YB-1were50(HSCORE≥200), accountingfor69.44%of the total, the lower expression of YB-1were22(HSCORE<200), accounting for30.56%of the total.60years of age or older were37,accounting for51.39%of the total, and younger than60were35, accountingfor48.61%of the total. The patients which clinical stage â… were15,accounting for20.83%of the total, clinical stage â…¡,there were47, accountingfor65.28%of the total, clinical stage â…¢,there were10, accounting for13.89%of the total. However, we observed no relationship between YB-1expressionand sex, age, stage.3. A lower DFS/OS was found in patients whose tissues hadhigher YB-1expression than whose lower(38.2m VS52.4m, P=0.013)/(44.4mVS57.3m,P=0.008).4.The clinical stage and with or withoutchemotherapy and the expression of YB-1are all of the factors affecting the prognosis of patients with rectal cancer. The age and sex of patients with rectalcancer was not statistically significant with the prognosis. Conclusion:1.Theexpression level of YB-1in rectal cancer were higher than rectal tubularadenoma and rectitis tissues.2.Shortend DFS and OS of patients whose tissuesoverexpressed of YB-1.3. The expression of YB-1may be one of theindependent prognosis factors of rectal cancer. |
