Objective:To investigate the effect of tibetan medicine Zuota on the pharmacokinetics of crocin-1in rats; to analyze the absorption, distribution and excretion of mercury in Tibetan medicine Zuota in rats.Methods:1. The plasma concentration of crocin was determined by HPLC after rats alternatively administered Zuota and crocin-1, and the pharmacokinetic parameters of crocin were calculated using a DAS Pharmacokinetic software. Thus the effect of Zuota on the pharmacokinetics of crocin-1was analyzed2. The mercury contents in serum, hepatic tissue, kidney tissue, urine and feces were determined by atomic fluorescence spectrometer to better understand the absorption, distribution and and excretion of Zuota in rats.Results:1. Crocin-1showed single compartment model in rats and the main parameters of pharmacokinetic were as follows:t1/2160min. Tmax87min, Cmax3.75μg/ml, AUC764μg.min/ml, CL0.019L/min/kg, V4.01L/kg, MRT141.92min.2. The pharmacokinetic parameters of crocin-1changed significantly after rats administered crocin-1and Zuota.The AUC,Cmax and MRT were significantly greater in rats than those of control rats (P<0.05), and the CL and Vd were significantly lower than those of control rats (P<0.05), and the AUC of crocin-1was greater in the21-day administration group than that of the7-day administration group.3. After rats were respectively administered Zuota for7days and21days, mercury levels were0.059±0.020and0.043±0.022mg/kg in serum,1.173±0.086and0.122±0.009mg/kg in hepatic tissue,5.937±1.421and7.296±1.115mg/kg in kidney,0.036±0.021and0.068±0.023mg/kg in the urine,42.148±6.421and95.876±12.119mg/kg in the feces. It can be seen that mercury content in serum was lower than that in kidney and liver. Mercury level increased in kidney, but decreased in hepatic tissue as administration period prolonged. A small amount of mercury was found in the urine, but a great quantity of mercury was excreted in the feces. Conclusion:1. After combined administration of Zuota, the absorption of crocin-1was significantly increased, and the clearance rate was significantly decreased in rats.2. Only a little mercury in Zuota was detected in rat plasma, and most of them were excreted to the environment with feces. The accumulation for mercury was observed in rat kidney after repetitive administration of Zuota for21days.
