Objective:The expression changes of P-selectin and it’s receptor L-selectinã€IL-8 and it’s receptor CXCR1/CXCR2ã€CD11a and it’s receptor ICAM-1 in asthma guinea pig Whether affect the ability of neutrophil adhesion and migration.Method:Useing our laboratory prophase method to building asthma model of guinea pigs. Thirty guinea male pigs with from eight-week-old to ten-week-old were randomly divided into three groups: control group,asthma group, dexamethasone treatment group(treatment group).Collecting bronchoalveolar lavage fluid(BALF) and blood for leucocyte classification count. Extracting guinea pig lung tissue for immunohistochemical experiment. By immunohistochemical detecting location and the expression of P-selectin and ICAM-1 on pulmonary vascular endothelial cells. Extracting the neutrophils of abdominal aorta blood for subsequent study which including the detections of protein related blood neutrophils expression of protein and m RNA level.Theprotein expression of CXCR1, CXCR2, L-selectin were detected by Western blot. The expression of the m RNA of CXCR1, CXCR2, L-selectin were tested by Q-PCR. Transwell Chambers detected the ability of neutrophil migration. Adhesion experiment detected the ability of neutrophil adhesion. The concentration of IL- 8 in serum and BALF were tested by enzyme linked immunosorbent assay(ELISA). Flow cytometry detected the expression levels of CD11 a on the blood neutrophil.Result:(1)The different groups of guinea pigs have different responses in the process of excitation: the guinea pigs of control group had no abnormal reaction. In the process of atomization inspire the guinea pigs of asthma group and treatment group shown typical symptoms of asthma which included cough, catch nose, nod breathing, shortness of breath, restlessness, etc. The symptoms of asthma guinea pigs were more obvious.(2)Leucocyte classification count results: The results of the percentage of neutrophils and eosinophils in BALF and blood of each group guinea pigs shown: asthma group compared with control group and treatment group,the former percentage was significantly increased,the differences were statistical significance(P < 0.01).(3)The results of the protein expression levels of CXCR1,CXCR2, L-selectin on the blood neutrophils of each group guinea pigs shown: asthma group compared with control group and treatment group,the former expression was significantly increased,the differences were statistical significance(P <0.01).(4)The results of the IL-8 expression level in serum and BALF of each group guinea pigs shown that: asthma group compared with control group and treatment group,the former expression was significantly increased,the differences were statistical significance(P < 0.01).(5)The results of the m RNA expression levels of CXCR1,CXCR2, L-selectin on the blood neutrophils of each group guinea pigs shown that: asthma group compared with control group and treatment group,the former expression was significantly increased,the differences were statistical significance(P < 0.01).(6)The results of the P-selectin and ICAM-1 expression and location on pulmonary vascular endothelial cells in lung tissue of each group guinea pigs shown that: asthma group compared with control group and treatment group,the former expression was significantly increased,the differences were statistical significance(P < 0.01).(7)The results of the migration ability of blood neutrophil in each group guinea pigs shown that: asthma group compared with control group and treatment group,the former migration ability was significantly increased,the differences were statistical significance(P < 0.01).(8)The results of the adhesion ability of blood neutrophil in each group guinea pigs shown that: asthma group compared with control group and treatment group,the former adhesion ability was significantly increased,the differences were statistical significance(P < 0.01).(9)The results of the expression levels of CD11 a on the blood neutrophil in eachgroup guinea pigs shown that: asthma group compared with control group and treatment group,the former adhesion ability was significantly increased,the differences were statistical significance(P < 0.01).Conclusion:(1)The expression of P-selectin and it’s receptor L-selectinã€IL-8 and it’s receptor CXCR1/CXCR2ã€CD11a and it’s receptor ICAM-1 were significantly increased in guinea pigs of asthma group.(2)The expression of L-selectinã€P-selectinã€CXCR1ã€CXCR2ã€IL-8ã€ICAM-1ã€CD11a were significantly up-regulated,which can strengthen the ability of neutrophil adhesion and migration.(3)Neutrophil increased in the airways of the asthma guinea pigs, which may be caused by the overexpression of these adhesion molecules and neutrophilic chemokines and their receptors. |
