Objective:To research the effect of Puerarin and cisplatin on proliferation and apoptosis of Non-small cell lung cancer A-549 cells in vitro, and the relationship between the expression of Bcl-2,Bax and PCNA protein, and SP can promote DDP chemotherapy effect on Non-small cell lung cancer A-549 cells.Methods:Use different drug concentrations of SP, DDP and SP+DDP effect on lung cancer A-549 cells.Use inverted microscope and optical microscopy observed the cells morphological changes.By MTT detect the drug dose and action time of the SP, DDP and DDP+SP effect on A549 cells. Use chemical assay immunohistochemistry detect the relationship between the expression of Bcl-2,Bax and PCNA protein in different action time and drug concentrations of SP, DDP and SP+DDP.Results:1. After different SP, DDP and SP+DDP concentrations effect on Non-small cell lung cancer A-549 cells, Use inverted microscope and optical microscopy observed the cells morphological changes.With the increase of drug concentration, the number of cells reduced, floating cells increased and the Cell morphology changed. There are also differences in number of cells and the cell morphology in different time period,the floating cells increased, the adherent cells reduced. After different SP, DDP and SP+DDP concentrations 48 hours effect on Non-small cell lung cancer A-549 cells, use HE staining observed the cells morphological changes. The cell in SP, DDP and SP+DDP occurs nucleus shrinkage, nuclear fragmentation, nuclear cytoplasm ratio decreased and cytoplasmic staining changed. With druge concentration increasde, the apoptotic cells corresponding increased.And SP+DDP group the number of apoptotic cells Greater than the SP and DDP group.2.With the increasing Concentrations of SP, DDP and SP+DDP effect on Non-small cell lung cancer A-549 cells, the OD value corresponding reduced, accordingly inhibition rate increased accordingly.So SP, DDP and SP+DDP in a concentration depend on the drug concentration. One drug with the growth of drug action time, OD value decreases, on the contrary, the inhibition rate increased. So, there have a time patience of SP, DDP and SP+DDP in action of Non-small cell lung cancer A-549 cells.3.The relationship between the expression of Bcl-2, Bax and PCNA protein in different SP, DDP and SP+DDP concentrations effect on Non-small cell lung cancer A-549 cells. With the increase of drug concentration and time growth, the Bcl-2 and PCNA protein reduced expression, but, the Bax protein increased expression.Conclusion:1. SP, DDP and SP+DDP inhibitory the proliferation of Non-small cell lung cancer A-549 cells, Promote apoptosis of Non-small cell lung cancer A-549 cells, and its make a dose- time-dependent manner.2. SP and DDP in a synergistic effect on Non-small cell lung cancer A-549 cells, reduce the rate of cell proliferation, promoting the rate of apoptosis in Non-small cell lung cancer A-549 cells.3. SP reduced the expression of Bcl-2 and PCNA protein, but, increased expressionof the Bax protein. SP can promote DDP chemotherapy effect on Non-small cell lung cancer A-549 cells.
