Objective: To study of morphological of gonadal axis changing of abnormal balgam syndrome with impotence disease rats model of Uyghur medicine,and changes in penile tissue NO-cGMP pathway key indicators,exploring the biological basis of abnormal balgam syndrome impotence model of Uyghur medicine.Methods: Selected SD rats of 70 mature male, taken out of ten for the control group(N),the other was the model group.To established animal models of abnormal balgam syndrome by spinach and coriander method into the cold humid environment.Weekly Watch biological characterization of rat,confirm Syndrome model.Experimental evaluation by APO erectile function experiment in rats, selected impotence model after 22 weeks.The impotence rats were randomly divided into model group(A1) and drug intervention group(A3).The non-impotence rats were randomly divided into model group(B1) and drug intervention group(B3).After three weeks,while observing biological characterization of the rats and evaluating erectile function.We detected the morphological changes of hypothalamus,pituitary and testis.Detected the morphological changes of penis by V.G staining and Masson staining.Immunohistochemistry detection of changes in penile tissue NO- c GMP pathway related factors.Results:(1)At the period of model,the change in the model consistent with the generalstate of Uygur medicine general characteristics of abnormal balgam,that is lying curled upto less,liking to get together,slow,dull hair,chaos,clear urine,pale stool the soft stool, darktongue,greasy tongue coating,slow weight gain,increased food intake,decreased waterintake,urine and stool output increased.After treatment with Yimusake,the symptoms were impoved.(2)At the period of model,erectile latency of model group was increased,and the number of erections was reduced.The incubation periods of straddle,insertion and ejaculation were longer than normal group,and the frequency of straddle,insertion and ejaculation is lesser than normal group.The rate of ED was 48.33%.After treatment with Yimusake,the incubation periods of straddle,insertion and ejaculation of A3,B3 were shorter than A1,B1;and the frequency of straddle,insertion and ejaculation of A3,B3 were more than A1,B1.(3)Morphological changes of gonad axis showed that A1,B1 group of rats hypothalamus, pituitary significant edema, A3, B3 group was significantly reduced edema.A1,B1 group of testicular germ cells of rats and the count of sperm significantly reduced,while A3,B3 group of rats values almost close to normal group.(4)The morphological changes in penile tissue showed that A1,B1 group of rats penile tissue significant edema.A3,B3 group was significantly reduced edema.V.G staining and Masson staining results showed,the ratio of smooth muscle/collagen fibers in penile tissue of A1,B1 group of rats was significantly lower than N group(P<0.05);A3,B3 group was higher than A1, B1 group(P<0.05),lower than the normal group(P<0.05).(5) Penile tissue immunohistochemistry showed,the expression of eNOS and nNOS of A1, B1 group were lower than normal group(P<0.05).A3,B3 group were higher than A1,B1 group(P < 0.05),Compared with normal group,and the expression of nNOS had significant difference(P<0.05),the expression of eNOS had no different(P>0.05).The expression of iNOS of A1,B1 group were higher than normal group(P<0.05).A3,B3 group was lower than A1,B1 group(P<0.05).Compared with normal group,A3 group was higher than normal group(P<0.05),while B3 group had no different(P>0.05).The expression of cGMP of A1,B1 group were lower than normal group(P<0.05).A3,B3 group was higher than A1,B1 group(P<0.05).Compared with normal group,A3 group was lower than normal group(P<0.05),while B3 group had no different(P>0.05).The expression of PDE5 were higher than normal group(P<0.05).A3,B3 group was lower than A1,B1 group(P<0.05).Compared with normal group,A3 group was higher than normal group(P<0.05),while B3 group had no different(P>0.05).Conclusion:(1)The change in the abnormal balgam syndrome and abnormal balgam syndrome with impotence disease rats model consistent with the general state of Uygur medicine general charactersistics of abnormal balgam,through spontaneous recover and Yimusake tablet treatment,the model is reliabity and stability.(2)The change of hypothalamus,pituitary and testicular of abnormal balgam syndrome and abnormal balgam syndrome with impotence disease rats model were boundaries nuclear transfer occurred was unclear,pathological changes in vacuolar and changes may be pathological basis of the disease syndrome gonadal axis hormones related disorders.(3)Reduced NO- cGMP pathway may be key to the development of abnormal balgam syndrome with impotence disease of Uygur medicine,which changes eNOS,nNOS,cGMP and other functional factors played a key role,but also the change of PDE5 was the key factor.(4)Yimusake for treatment of abnormal balgam syndrome with impotence disease of Uygur medicine may promote gonadal axis and pathological changes related to the role of repairing.(5)Yimusake for treatment of abnormal balgam syndrome with impotence disease of Uygur medicine may active NO- c GMP pathway,increased eNOS,nNOS,cGMP and other related key factors.(6)Yimusake for treatment of abnormal balgam syndrome with impotence disease of Uygur medicine may be the many targets,it can effectively inhibit the expression of PDE5,improving treatment effect.
