Objective:Based on the Uighur medical theory to establish abnormal Balgam syndrome andimpotence rat model, determine its effections to neurotransmitter and rate-limitingenzymes expressions in penis erection related signaling pathways and on this basis toinvestigate molecular mechanisms of the abnormal Balgam induced impotence.Methods:100male SD rats with normal sexual function were randomly selected anddivided into normal group (N)10, rest of them for the model group. Normal grouprats were reared with ordinary feed in normal rearing environment. Model group ratswere treated with wet cold feed (ordinary feed mixined with coriander and spinach)20weeks under the conditions of the wet and cold established abnormal Balgamsyndrome rat model. Through APO erectile function experiments, screening theabnormal Balgam syndrome indued impotence rats from the abnormal Balgamsyndrome rat model, established abnormal Balgam indued impotence rats model.Abnormal Balgam syndrome rat model were screening into several groups: Balgamsyndrome indued impotence model rats were divided into impotence disease group(A1), natural recovery group (A2), treatment group (A3); syndromes group (B1),natural recovery group (B2), treatment group (B3) and A3, B3group rats were treatedwith Yimusake two weeks. Through Western blot assay detecting expression of eNOS,nNOS, iNOS, VIP, CGRP in penile tissue from each group rats.Results:1. Western-blot results show that, A1group and group A2rat penile tissue eNOSexpression level to be compared with the N group has been significantly reduced(P<0.01, P<0.01); the group A3amount of eNOS expression compared with group A1,A2group has been significantly increased (P<0.01, P<0.01), the A2group comparedwith the A1group no significant difference (P>0.05); B3expression of eNOScompared with B1group has been significantly increased (P<0.05), compared with theB2group showed no significant difference (P>0.05); group B2and B1groups showedno significant difference (P>0.05).2. Each model group penile tissue nNOS expression level compare with N group,A1, A2group nNOS expression level has been significantly lower (P<0.01, P<0.01);A3group compare with the A1, A2group has been significantly increased (P<0.01, P<0.01), the A2group compare with the A1group no significant difference (P>0.05);B3group compare with B1, B2group comparison has been significantly increased(P<0.01, P<0.05); B2and B1group showed no significant difference (P>0.05).3. iNOS expression in the N group compared with other groups, group A1, A2group was significantly higher (P <0.01, P<0.01); the A3group compared with the A1group had significantly lower (P<0.05), compared with the group A2no significantdifference (P>0.05); the B3group compared with B1, B2group has been significantlyreduced (P <0.05, P<0.05).4. Group N rats penile tissue the VIP expression levels compared with A1groupand group A2has been significantly reduced (P <0.01, P <0.01); the A3groupcompared with group A1, A2group comparison with significantly increased (P <0.01,P <0.01); the B3group compared with B1, B2group has been significantly increased(P <0.01, P <0.01); group B2compared with group B1significant difference (P>0.05).5. The N group rats penile tissue expression of CGRP comparison with eachgroup showed no significant difference (P>0.05).Conclusions:1. The abnormal Balgam induced impotence rat model of penile tissue eNOS,nNOS, iNOS expression levels compared with normal group had significantdifference; eNOS, nNOS, iNOS is the major rate-limiting enzyme of the NO-cGMPsignaling pathway, and its content may affectNO-cGMP signaling pathway, furtheraffecting erectile function.2. Abnormal Balgam induced impotence rat model of penile tissue VIPexpression level compared with normal group had significant difference,CGRPexpression show no ignificant difference. VIP is VIP-cAMP main neurotransmitter inerectile signaling pathway, and its content changes may affect erectile function.However the CGRP expression and its signal transduction pathway CGRP-cAMP maybe unaffected.3. While model rat treated with Uygur medicine, eNOS, nNOS, iNOS, VIPexpression in penile tissue has a significant reversal. Suggesting that eNOS, nNOS,iNOS, VIP may be crucial to abnormal Balgam induced impotence moleculemolecular mechanisms research.
