The Correlation Analysis Between The Traditional Chinese Medicine ZHENG And The Expression Of Plasma Prognostic MiRNA In Non-resectable Pancreatic Cancer

ObjectiveTo discuss the correlation between the traditional Chinese medicine ZHENG and the prognostic miRNA in non-resectable pancreatic cancer patients in order to pave the way for the Molecular prediction model in traditional Chinese medicine ZHENG.MethodsPart I:To review the advance of traditional Chinese medicine treatment in non-resectable pancreatic cancer, in order to follow the step of the research in the same field all over the world.Part II:Screen the prognostic miRNA with the technology of miRNA sequencing. Choose the candidate miRNAs for the step of validation which is tested by the real-time PCR technology on the basis of Fold change value and found the predictive prognostic miRNA in non-resectable pancreatic cancer patients.Part III:retrospective analysis of the correlation between traditional Chinese medicine ZHENG and the overall survival in pancreatic cancer patients. to discuss the correlation between traditional Chinese medicine ZHENG and prognostic miRNAResultsPart I:we get the information of the reaearch in the same field, and know our issue thoroughly with the review of traditional Chinese medicine treatment in non-resectable pancreatic cancer.Part II:we have screened many prognostic miRNAs with the technology of miRNA sequencing, and choosed ten candidate miRNAs for the validation process on the basis of Fold change value, after the testing with the technology of real-time PCR, we have got two true positive miRNA which is correlated with the prognosis of non-resectable pancreatic cancer patients. The overall survival between the miR-19b low expression patients and the miR-19b high expression patients had a significantly differences (P<0.05). The median survival time is 7.86±1.29 and 5.12±0.59 months(P=0.106), the overall survival between the miR-122 low expression patients and the miR-122 high expression patients also had a significantly differences (P<0.05). The median survival time is 5.67±2.56 and 9.25±4.85 months (P=0.000), so we think that the miR-19b and miR-122 could be served as the prognostic miRNA in non-resectable pancreatic cancer patients.Part Ⅲ:we have made the survival curve in the three group cancer patients(PI-XU, XUE-YU, SHI-RE), and we have also found that the SHI-RE group has the better suivival, there is no difference between the PI-XU group and XUE-YU group. The overall survival time is 7.86±1.29,5.12±0.59 and 11.79±1.27 months (P=0.001). The different expression of prognostic miRNA contributes to the significantly differences of overall survival in the three groups. So we compared the expression of miR-19b and miR-122 in the three groups, and we have found that the expression of two miRNAs is higher in SHI-RE group. The expression of miR-19b are 0.7246±0.1466、 0.6999±0.2861 and 0.9998±0.4556 (P<0.05); and the expression of miR-122 are 0.4627±0.2414,0.4524±0.2446 and 1.1185±0.7077 (P<0.05)ConclusionMiR-19b and miR-122 could be served as prognostic miRNA in non-resectable pancreatic cancer patients, and there exist the correlation between the traditional Chinese medicine ZHENG and the prognostic miRNA.
