Objective:To study the different extracts of Ferula ferulaeoides in vitro inhibition of gastric cancer,and to study the preparation technology of active site,study its active site to study the chemical composition, and provide scientific basis for further development and utilization of the traditional Uighur medicine.Methods:1.Through different polar solvent to extract more ferula ferulaeoides, and using MTT test,the cell apoptosis HOCHEST33258 staining experiment, flow cytometry and Annexin V-FITC/PI testing different extracts in gastric cancer cell line SGC-7901 growth;2.To extract rate and the inhibition on gastric cancer cell SGC-7901 as an index, using the single factor and orthogonal test L9 (34), the method of combining for more than an umbrella ferulic chloroform in vitro anti cancer of the stomach to optimize the extraction process of active site; 3.By using GC-MS, HPLC method for multiple f.ferulaeoides inhibiting gastric cancer study on fingerprint active site;Results:1.Umbrella volatile asafetida oil, aqueous extract,95% ethanol extracts, petroleum ether extract, chloroform extract, ethyl acetate extract had inhibitory effect on gastric cancer SGC-7901 cells, the inhibitory effect of chloroform extract and ethyl acetate extract for SGC-7901 gastric cancer cell proliferation is obvious; and the inhibitory action of chloroform extract is the strongest,compared with the control group obviously, and with the drug concen tration increased,the inhibition rate is higher the concentration gradient dependent;2.More umbrella ferulic chloroform in vitro inhibition of gastric cancer the best extraction process of active site:in f.ferulaeoides powder for the extraction of objects, using chloroform as extraction solvent Ferula ferulaeoides,Ferula ferulaeoides powder 10 mesh sieving, adding 8 times volume fractions of chloroform extration,,60℃ extraction, extraction for 3 times, each time 60 min; 3.Established the method analysis of Ferula ferulaeoides chloroform in vitro anti gastric cancer active site GC-MS, HPLC fingerprinting, GC-MS fingerprinting was established on 28 common peaks, monoterpene composition 3, sesquiterpenes have 19, aliphatic composition 3, aromatic constituents of 3, a total of 10 batches of samples the above components are the majority of the total composition of 92%, its similarity to reach more than 0.9; the HPLC fingerprints and establish 13 common peaks,10 batches of samples a total of more than 58% total peak components composition,the similarity to reach more than 0.9; Conclusion:1.Tentatively confirmed Ferula ferulaeoides chloroform extract had inhibiting gastric cancer cell SGC-7901 interaction potential, further still needs to be verified by animal cells in vivo;2.The 3 batch of sample preparation the results showed that the anti gastric cancer in vitro active site of Ferula ferulaeoides chloroform extraction technology is reasonable, stable. The extract rate more than 19.8%;and pharmacodynamics in vitro experiment showed that the 3 batch of the pharmacological activity and activity experiments in vitro active site preparation of samples for the screening of chloroform extract activity was not different between the description, preparation process opyimized and achieved the expected results.3.Established by GC-MS and HPLC fingerprint method is rapid, simple, with precision, repeatability, stability good, in clinic, can be used as the quality control method of Ferula ferulaeoides chloroform in the active site,for the development of further innovation of drugs, to formulate a scientific, reasonable, strict quality control standards and evaluation system established foundation.
