Cistanche Deserticola is the stem of Cistanche deserticola Y.C.Ma.or Cistanche tubulosa(Schrenk) R.Wight.It has the functions of enforcing kidney,benefiting essence and blood,including cathartic and anti-aged action with salty property and warm flavor.As the original plant of Cistanche Deserticola,Cistanche tubulosa (Schrenk) R.Wight.has been used for treating diseases with a history of over one thousand years in china.As a result of its good biological activity and sustainability, more and more researchers,both at home and abroad,have begun to focus on the research and development of Cistanche Deserticola in the field of drugs as well as functional food.In order to get a further usage of the product,this article discussed a systematic research on Cistanche tubulosa(Schrenk) R.Wight.through five aspects as follows:First,the article summarizes the research and development of Cistanche Deserticola on its plant resources,chemical constituents,pharmacological reaction and produce technology in detail and analyzed its market prospects through literature and investigation.Of the 22 kinds of Cistanche Deserticola all over the world,there are 6 kinds in China and 5 of them live in desert,including:Citanche deserticola Y.C. Ma,Cistanche tubulosa(Schrenk) R.Wight,Cistanche Salsa(C.A.Mey) G Beck, Cistanche Salsa(C.A.Mey) G.Beck varalbiflora P.F.Tu et C.Lou.The chemical constituents includes:phenylethanoid,irodoid glycosides,lignanoside and various amino-acid.Cistanche tubulosa(Schrenk) R.Wight herb has the functions of enforcing kidney,benefiting blood and cathartic as recorded in CP2005.The research of modern pharmacology indicates that the herb can enforce the activities of immunity and male hormone,cathartic,repair the damaged cell,anti-oxidant and anti-aged,also can improve the memory and protect the ischemia-reperfusion region,and indicate for the treatment of leukemia,septic shock.Lots of literatures have reported the technical process of extracting from the herb the indexed constituents such as phenylethanoid, betaine and poly- saccharide.Second,study on determination of the indexed constituents including Echinacoside and Acteoside in herb and extracting from Cistanche tubulosa(Schrenk) R.Wight.It is by the method of ultrasound in methanol that Cistanche tubulosa (Schrenk) R.Wight is extracted through the optimized process.The detected conditions are as follows:mobile phase:methanol-acetonitrile-l%acetate(15:10:75), detected wavelength:334nm,flow rate:0.8 ml·min-1,column temperature:30℃.By this method,both Echinacoside and Acteoside can be detected with good separability and less impurity peak within 28 minutes.Furthermore,the study on stability shows that Echinacoside and Acteoside can be kept and protected from light at room temperature for 12 hours,or at refrigerator temperature for 1 week.So it concludes that the method is simple,accurate and applicable for the determination of Cistanche tubulosa(Schrenk) R.Wight.Third,Cistanche tubulosa(Schrenk) R.Wight herbs got from market are extracted by methanol and studied on the fingerprint of the extracted fluid by re-HPLC.14 chromatogram peaks from Cistanche tubulosa(Schrenk) R.Wight are well isolated and the chemical data which can indicate the quality of the herb are obtained in this way.The herb samples are divided into 3 grades including high quality,common quality and low quality through hierarchical clustering analysis.And through the count of similarity,the result also shows that the similarity of high quality,common quality and low quality samples are 0.90~1.00,0.70~0.89 and below 0.70 respectively.So the identification,classification and grade of the unknown sample can be analyzed through the similarity between tested and standard samples.The study established the method for quality evaluation of herbs,which is especially practical for the product evaluation in factories.Forth,combined with the feasibility in factory,the extracted technical process was optimized by orthogonal design L9(34) The dry extracting yield of Echinacoside and Acteoside were the referent indexes.The results showed that ethanol was the proper extract solvent and the remarkable influence factor was extracted time.The best extracted processes were:6 times 50%ethanol and extracting 2h for three times.The dry extract yield was 18%and the total content of both Echinacoside and Acteoside was 9.72%.The optimal extraction technology is feasible with high content of Echinacoside and Acteoside,and provides the basis for the actual production.At Last,after a comparison between the 6 kinds of macroporous resin from static absorption to desorption rate,D1400 macroporous absorption resin was selected for isolating and purifying phenylethanoid glycosides from Cistanche tubulosa(Schrenk) R.Wight.for its good isolation and purification effect.Some impurities can be get rid of through elution by 5%~10%ethanol firstly and then eluted by 50%ethanol.The product which attained in 50%ethanol was determined after decompressing concentration and vacuum dry.The content of phenylethanoid was 32.53%in the treated powder and meet with the standard in international market.The study provided the reference for collecting and purification of phenylethanoid in Cistanche tubulosa(Schrenk) R.Wight.herb.
