| Purpose: Through the observation in total of Yang-deficiency syndrome complicated with myocardial ischemia rats model, to investigate the protective effect of Guan Xin Fu Han Granule on the composite of myocardial ischemic injury in rat animal model.Method: With continuous oral administration of 14 days DEX replication animal model of Yang deficiency, based on the ligation replication of coronary artery in rats model of myocardial ischemia, to observe the Guan Xin Fu Han granule drug effects on myocardial ischemic model.Result:(1)The influence on the animal model of myocardial ischemic injury(electrophysiological method) shows that the Guan Xin Fu Han granule to offset millivolt values of Medicine animal ECG ST segment was significantly lower than the same period in model group(P<0.05). It shows that Guan Xin Fu Han granule has obvious improvement at the degree of myocardial ischemia injury in dexamethasone Yang-deficiency syndrome model of myocardial ischemia rats.(2) Observing the effects on myocardial ischemia degree of Guan Xin Fu Han Granule on coronary artery ligation model. The value of total integral necrosis area under the ligation point was used as observation indexes in the method of the histological morphology by microscope. The results show that myocardial injury scores of Guan Xin Fu Han granule animal group are lower than those in the model group, which proved that Guan Xin Fu Han granule has certain protective effects on dexamethasone Yang-deficiency syndrome with myocardial ischemia injury.(3) Observing the effects of Guan Xin Fu Han granule on dexamethasone Yang-deficiency complicated myocardial ischemia injury animal model, three myocardial enzymes were used as index by the methods of spectrophotometry. The results : compared with the model group, the activity of LDH was significantly reduced in Guan Xin Fu Han granule high dose group and positive control group of animal serum(P<0.05), and the CK activity decreased obviously too separated in low, high dose groups of Guan Xin Fu Han granule,and even in positive control group(P<0.05), and the levels of AST reduced significantly in three groups(P<0.01). All showed that Guan Xin Fu Han granule has certain protective effect on dexamethasone of myocardial ischemia complicated with Yang-deficiency syndrome rat model of myocardial ischemia damage.(4)Using ELISA methods, ischemia modified albumin and cardiac troponin I were used as the index to observe the effects of Guan Xin Fu Han granule on myocardial damage degree of myocardial ischemia with dexamethasone Yang-deficiency syndrome animal model. Results showed that IMA content decreased(P<0.05), and cTnI decreased significantly too(P<0.05, P<0.01, P<0.001) in Guan Xin Fu Han administration group, suggesting that Guan Xin Fu Han granule has determinate protective effect on myocardial injury.(5) Effects of Guan Xin Fu Han granule on the whole state of myocardial ischemia with dexamethasone Yang-deficiency animal model. Results showed that the animal appeared to fur-haggard, standing, lying to themselves, reduced shoulder arch, slow-moving, dispirited after dexamethasone intragastric administration, cold and bleak tail, back temperature drop, dorsal edema, and decreased body weight. It showed that large dose of dexamethasone by oral administration can cause a series of symptoms of Yang-deficiency, for example, the whole animal shape change, But the body weight of administration group animal had no difference compared with the model group, which implied that Guan Xin Fu Han granule has no effect on the body weight of Yang-deficiency animal model.Conclusion: Guan Xin Fu Han granule(GFG) can lessen the degree of myocardial injury in animal models, and has a protective effect on Yang-deficiency syndrome complicated with myocardial ischemia animal myocardial cell injury. |
