Traditional Chinese medicine containing toxic alkaloids such as Strychnos Alkaloids, because of its safety, are easy to take in inappropriate treatment dose or preparation methods and cause poisoning. At the same time, due to its important medicinal value, Strychnos Alkaloids has long been the attention of the researchers and research work has been involved in the extraction and preparation technology, formulation and efficacy, pharmacology and toxicology, chemical components, pharmacokinetic, detection technology, and many other aspects. However, rare Strychnos Alkaloids metabolism researches were reported. Strychnos Alkaloids metabolism in vivo, in fact, research is of great significance, not only can easily poisoning cases inspection, and monitoring the concentration of this kind of drug metabolites in the body, so as to ensure the safety and rationality of use. So this paper will establish a reliable method to analyse the metabolism of Strychnos Alkaloidsin vivo and quantitative analysis of metabolites in blood.Performance liquid chromatography (HPLC) and high resolution mass (HRMS) is a liquid separation combined with high resolution mass spectrometry detection of new analytical instruments. With the development of HRMS by large equipment for desktop instrument, its development also more rapidly in recent years, has become an commonly used laboratory equipment. Using ultra high resolution, HRMS compounds can be accurately measured in a reliable way of precise mass than nuclear, and can distinguish the complex background of mass closer to the impurity and the total outflow components, trace compounds in complex matrix analysis ability. Using high resolution scanning mode is not missing information of unknown compounds such as metabolites, can is a kind of target or targets of drug toxicology screening method, get more and more application in the field of toxicological analysis.This research is based on the technology platform of LC-HRMS and in vivo metabolism of the Strychnos Alkaloids, specific work summarized as follows:The LC-HRMS technology used for Strychnos Alkaloids metabolites identified in the urine of rats method, choose the solid phase extraction method for sample pretreatment, and use the ultra-high performance liquid/linear ion trap-Orbitrap mass spectrometry (UHPLC)/LTQ Orbitrap-MS technology are analyzed.26 kinds of strychnine in rat urine metabolites and 23 species of strychnine metabolites were identified; First deduce 25 kinds of metabolic product and four in vivo metabolic pathways. This study not only more comprehensive expounds the rats in the metabolic characteristics of Strychnos Alkaloids, also to work in the field of toxicology screening and metabolite identification provides a certain reference value.Established at the same time qualitative and quantitative analysis of two kinds of Strychnos Alkaloidsin human blood (strychnine and ning) and its metabolites of LC-HRMS analysis method, and has carried on the investigation and verification of the methodology. The results show that the method meets the requirement of biological analysis.
