Qingyi Granule is an empirical formula optimized from Qingyi Decoction.It has the effects of relieving pain,dredging and purging heat,which can be used for clinical treatment of chronic pancreatitis.There is currently no research on the medicinal material basis of this prescription.In order to promote the modernization and internationalization of Qingyi Granules,and provide scientificity in clinical use,the research on the medicinal material basis of Qingyi Granules is imminent.This article intends to combine the high resolution and high resolution to explain the medicinal material basis of Qingyi Granules from two aspects:the composition analysis of Qingyi Granules and the metabolism study of Qingyi Granules in rats.In this study,by optimizing the extraction conditions and liquid phase conditions,the HPLC-UV-HRMS/MS method was established.Compounds were separated on Agilent Zorbax SB C18 column(150×4.6 mm,5μm),using acetonitrile and 0.3%acetic acid solution in water as mobile phase for gradient elution,with a flow rate of 0.5 m L/min.Qingyi Granules exact was analyzed by MS and MS~n full scan in both positive and negative ionization modes.According to chromatographic and mass spectrometry behaviors,as well as comprehensive literature,reference materials,databases and other related data,the chemical components contained in Qingyi Granules were identified.A total of 201 compounds were identified from Qingyi Granules extract by HPLC-UV-HRMS/MS method,of which 127 were identified in positive ionization mode and 124 in negative ionization mode.There were 76 flavonoids,21alkaloids,35 terpenoids,14 amino acids,21 phenylpropanoids,7 anthraquinones,15 organic acid compounds and 13 other compounds.The method was fast and sensitive,realized the simultaneous identification of multiple compounds,and lays a solid foundation for the material basis research and quality control of Qingyi Granules.The metabolism of Qingyi Granules in rats was also investigated by HPLC-HRMS/MS.Qingyi Granules were administered intragastrically to rats at a dose of 900 mg/kg,and the corresponding urine and feces were collected.The results showed that 31 metabolites and 24prototype components were detected in urine,and 5 metabolites and 10 prototype components were identified in rat feces.Metabolites detected in urine are produced through phase I and phase II metabolism;metabolites in feces were mainly produced through phase I metabolism,and also partially produced by methylation.The metabolism study of compound Chinese medicine in vivo was performed through the Target-Group-Change strategy and HPLC-HRMS/MS method,this provides ideas and basis for the research on the metabolism of compound Chinese medicine,and also promotes the research on the medicinal material basis of Qingyi Granules.
