Objective This study was to retrospectively evaluate the efficacy and safety of raltitrexed as second-line treatment for advanced gastric carcinoma.Methods Datas were retrospectively collected derived from patients with pathologically proved advanced gastric cancer who have received second-line chemotherapy in the Affiliated Cancer Hospital of Guangxi Medical University during the period from June,2010 to November,2014. The treated with raltitrexed chemotherapy patients as the observation group, age, ECOG score and metastasis of organ and the number ratio factor, according to the ratio of 1:1 during the same period in the Affiliated Cancer Hospital of Guangxi Medical University diagnosed and accept not containing raltitrexed second-line chemotherapy patients as control group, analysis the curative effect and adverse reaction between the two groups. Objective efficacy was evaluated by using RECIST 1.1 standard, and the grade of adverse reactions was classified by the toxicity grade (3) of the National Cancer Institute. PFS, OS survival curves were described by Meier-Kaplan.Results 77cases were included:37 cases received raltitrexed for plug scheme(observation group),40 cases received contains no raltitrexed for plug scheme(controlled group). There were no significant differences in age, gender, ECOG score and metastasis of organ between the two groups (P> 0.05).In observation group, CRO cases, PR4 cases, SD20 cases, PD13 cases, DCR was 64.86%, ORR was10.81%.In controlled group CRO cases, PR3cases,SD24cases,PD13cases, DCR was 67.50%,ORR was7.50%.The median PFS was 3.35 months and 3.54 months (P=0.780)and OS was 4.39 months and 4.96 months (P=0.349) in observation group and controlled group. Two groups of the main adverse reactions of the white blood cells and neutropenia, nausea, vomiting, transaminase lifts.Related adverse reaction is â… -â…¡, â…¢-â…£ white blood cells and neutropenia in the observation group 2 cases (5.4%),2 cases in the control group (5.0%), no fever granulocytopenia correlation.In the observation group4 cases â… -â…¡ cardiac toxicity reaction, the control group in 4 cases of â… -â…¡ degree of peripheral nerve toxicity.Conclusions For the second-line chemotherapy of advanced gastric cancer patients,raltitrexed treatment can achieve efficacy, side effects can be tolerated, which has a broad prospect in clinical treatment.
