| Objective To investigate the silent information regulator 2 related enzyme 1(SIRT1) in osteoarthritis(OA) expression in synovial tissue and cells in correlation.Methods â‘ OA group: Synovial tissue from osteoarthritis of the knee due to total knee arthroplasty at department of bone tumor and joint from September 2013 to June 2014 at the First Affiliated Hospital of Anhui Medical University. A total of 20 patients, 10 males, 10 females, aged between 62-78 years, mean age 72.3 years.The inclusion criteria:In compliance with the ACR recommended in 2001 OA diagnostic criteria;â‘¡Normal control group: Normal control groups: normal synovial tissue from patients achieved because of acute trauma or tibial plateau fracture surgery. A total of 10 patients, 6 males, 4 females, aged between 19-42 years, mean age 34.2 years.The inclusion criteria:Postoperative pathological diagnosis with no joint lesions.After in vitro specimen placed in a sterile state decentralization equipped sterile saline specimen box, sent to the laboratory within 2 h were isolated and cultured synovial cells.Toluidine blue staining of histological examination of synovial cells, synovial cells of SIRT1 expression detected by immunohistochemistry to detect SIRT1 expression and distribution, expression was detected by Western blot in the synovial tissue and cells of SIRT1.Results 1ã€Toluidine blue staining revealed that,most of synovial cells were spindle poles slender cytoplasmic processes, end was connected with the adjacent cells, interwoven into a network, network can be seen between the few small stellated or polygonal cells. There were no significant differences between normal morphology and OA synovial cell group. 2ã€Immunocytochemistry showed that:In the normal group and OA synovial cell group were seen SIRT1 expression. Extensively in the cytoplasm was brown,the nucleus and the cell membrane was no obvious jaundice,OA group in comparison to normal staining intensity was significantly lower, the difference was statistically significant(t=20.208,P﹤0.01). 3ã€Western blot analysis showed that:in OA synovial tissues, SIRT1 expression is lower than the normal group,gray value normal group and OA group were statistically analyzed and compared, the difference was statistically significant(t=7.664,P<0.01),Gray value normal and OA synovial cell group comparisons were statistically significant by statistical analysis(t=8.619,P﹤0.01).Conclusion The low expression of SIRT1 may play an essential role in OA progression, and the research provides a new theoretical basis for SIRT1 as therapeutic target in OA treatment... |
