Eucommia ulmoides seed-draff extract (ESE) could promote the proliferation and differentiation of osteoblasts, restrain bone resorption, and increase the BMD of PMOP model rats. The aim of our present study is to study the anti-osteoporosis mechanism of ESE and explore the internal relationship among reinforce the healthy qi and secure the root, tonify the kidney and strengthen bone in traditional Chinese medicine theory.We found that ESE administration could significantly enhance the activity of spleen lymphocyte and peritoneal macrophages in vitro, but had no effect on the activity of thymocyte by MTT assay. The PMOP model was made by bilateral ovariecshangctomy. The experimental results in vivo showed that treatment with ESE could increase the spleen index of PMOP model rats, obviously up-regulate CD4+ T lymphocytes level, down-regulate CD8+ T lymphocytes level, increase CD4+/CD8+, and increase the Foxp3 expression of CD4+CD25+Foxp3+Treg cells by flow cytometry. This phenomenon was well correlated with significant suppression of the overproduction of inflammation cytokines (TNF-α and IL-6) versus the osteoporotic rats with water treatment. Meanwhile, immunoturbidimetry and ELISA experiments results showed that the release of immunoglobulin (lgG and lgM), complement (C3 and C4) and IGF-1 were profoundly increased. In conclusion, ESE could regulate the function of immunoprotection and immunosuppression of PMOP model rats immunity system through the organismal, cellular and molecular levels experiments. This research implied that immunity regulation might be one of the main mechanisms of ESE anti-osteoporosis ativity. |
