| Objective : ã€To investigate the estrogen receptor(ER) progesterone receptor(PR) and growth hormone receptor(GHR) expression and significance of endometrial tissue in patients with intrauterine,adhesions which provide a theoretical reference for the postopera-tive use of growth hormones, estrogen and progesterone to prevent further adhesions in IUA patients under Transcervical Resection Of Adhesions(TCRA)Material and Methods:1.GroupingThe study group:Take the 28 patients who were diagnosed with intrauterine adhesion by hysteroscopy at the Second Affiliated Hospital of Anhui Medical University from November 2013 to May 2014, aged 20 to 35 years, mean age 29.62 ± 2.52 year old.The control group:22 patients at the same period for other reasons(infertility, uterine mediastinum or IUD incarcerated) who were diagnosed with non- intrauterine adhesions as the control group, aged 21 to 40 years, mean age 26.80 ± 3.34 year old. And all these were with history of uterine operations before.2. Methods2.1 Specimen collection.Research group: patients underwent hysteroscopy diagnosed as intrauterine adhesions, hysteroscopy or in the process of vaginal dissection hysteroscopy operation, use forceps to take the appropriate endometrial tissue(mass of about 20 mg) as specimens in the remaining normal endometrial tissue as needed.Control group: patients due to other reasons for non-hysteroscopy confirmed intrauterine adhesions and have had a previous history of uterine operations, such as uterine mediastinum, infertility, or IUD incarcerated, take some endomrtrial tissue as needed2.2 Experimental methods. Immunohistochemical Max Vision two-step method and real-time fluorescence quantitative PCR(q RT-PCR) are used for detection of GHR protein, ER protein, PR proteins and GHR m RNA, ER m RNA, PR m RNA in endometrial tissue both in group IUA(Study Group) and non IUA group(control group).Results:1.Immunohistochemical Max Vision two-step method to detect ER protein, PR protein, GHR protein both in study and control groups; q RT-PCR detect ER m RNA, PR m RNA, GHR m RNA expression both in study group and the control group; two experimental method for detecting GHR, ER expression in both groups had statistically significance(P <0.05), PR expression was not statistically significant(P in both groups> 0.05).2. Immunohistochemical Max Vision two-step method to detect the ER protein, PR protein, GHR protein both in endometrial glands and endometrial, stroma,the difference in the two parts was statistically significant(P <0.05).Conclusions1. The expression of GHR in patients with IUA is lower than those of non-IUA, and its expression is more in endometrial glands, less in endometrial stroma,which provide a theoretical reference for the use of hormone in IUA patients after TCRA.2. The expression of ER in endometrial tissue of study group was higher than in the control group, the expression of PR was no different between the two groups; ER and PR expression are more in endometrial glands, expressed less or even no expression in endometrial stromal; That provide a certain theory reference for clinical use of estrogen and progesterone to IUA patients after TCRA. |
