Objective: The thesis aims to analyze the relationship between TGF-β1 and MDR1, PTEN, TOPOII, ERCC1, LIVIN drug-resistance genes through the detection of TGF-β1 in epithelial ovarian cancer, and screen the related drug-resistance genes with TGF-β1 and make TGF-β1 tumor drug resistance molecular markers and novel targets for treatment through multiple gene joint detection which is used to guide clinical drug more objectively and comprehensively in order to reduce resistance, increase the survival rate of the patients.Methods: The essay detects the situations of expression of TGF-β1 group(50 cases) in epithelial ovarian cancer, benign epithelial ovarian tumor tissue(20 cases) and normal ovarian tissue(20 cases) respectively by means of immunohistochemical staining, statistical analyses of count data is achieved by chi-square test SPSS19.0 Statistical Software, and Spearman Rank Correlation in order that the differences have statistical significance with P < 0.05.Results: 1 The Expression of TGF-β1 in Ovarian Epithelial CancerTGF-β1 positive expression is mainly located in cytoplasm, which is shown in tan grains dyeing.There are 4 cases of TGF-β1 showing the positive expression within the given 20 normal ovarian groups, so the rate of positive expression is 20%(4/20); There are 8 cases of TGF-β1 showing the positive expression within the 20 benign ovarian tumor groups, so the rate of positive expression is 40%(8/20); There are 38 cases of TGF-β1 showing the positive expression within 50 ovarian epithelial cancer groups, so the rate of positive expression is 76%(38/50) which is growing gradually. The comparative differences have statistical significance(P<0.05) in the comparison between the TGF-β1 expression aspect of epithelial ovarian cancer and normal ovarian tissue. The comparative differences have statistical significant(P < 0.05) in the comparison between the TGF-β1 expression aspect of epithelial ovarian cancer and benign ovarian tumor. The differences have no statistical significance(P> 0.05) in the comparison between the TGF-β1 expression aspect of normal ovarian tissue and benign ovarian tumor. 2 The Relationship between TGF-β1 Expression in Ovarian Cancer Tissue and the Clinical Pathological Features as Shown in Table 2 2.1 AgeThe positive expression rate of epithelial ovarian carcinoma tissue is 78.5%(22/28) in the age of 50 or over, the positive expression rate of epithelial ovarian carcinoma tissue was 72.7%(16/22) in the age<50, and the differences have no statistical significance between them.(χ2=0.231, P > 0.05). 2.2 By StagesTGF-β1 positive expression rate of 88.2%(30/34) in â…¢- â…£stage of epithelial ovarian cancer is higher than the positive expression rate of 50.0%(8/16) in stage of â… - â…¡, so there is statistical significance in the differences between them(χ2=8.720, P<0.05). 2.3 Histological TypesThe positive expression rate of TGF-β1is 79.2%(19/24) in serous cystadenocarcinoma, 68.8%(11/16) in mucinous cystadenocarcinoma, 80.0%(8/10) in endometrioid carcinoma, so there is statistical significance in the differences among them(χ2 =0.739, P>0.05); 2.4 The Histopathological DifferentiationThe positive expression rate of TGF-β1is 66.7%(16/24) in high school, 84.6%(22/26) in low school in epithelial ovarian cancer.There is statistical significance in the differences between them(χ2=2.204 P>0.05). 2.5 Lymphatic MetastasisThe positive expression rate of TGF-β1 is 81.3%(26/32), and the positive expression of TGF-β1 is 66.7%(12/18), which there is statistical significance in the differences between them(χ2 =0.663, P>0.05); The expression of TGF-β1 has no business with age, histological type, histopathological grading and lymphatic metastasis of the patients. 3 The Relationship between the Expression of TGF-β1 and MDR1, PTEN, TOPOII, ERCC1, LIVIN Drug-resistance Related Genes:The expression between TGF-β1 and MDR1 has a positive correlation(r = 0.322, P < 0.05);TGF-β1 and PTEN has a negative correlation(r =-0.668, P < 0.05);the expression of TGF-β1 and TOPOII has no correlation(r =-0.506, P = 0.078), TGF-β1 has no correlation with ERCC1 expression(r =-0.037, P = 0.636), and the expression between TGF-β1 and LIVIN has no correlation(r =-0.801, P = 0.21).Conclusion:1 The positive expression level of TGF-β1 in epithelial ovarian cancer is significantly higher than benign ovarian tumor and normal ovarian tissue. From normal ovarian tissue to benign tumors and epithelial ovarian cancer, the expression of TGF-β1 shows a trend of rising which the differences of the three groups are apparent, showing TGF-β1 plays an important role in the process of from benign to deterioration in ovarian tissue, so TGF-β1 can be used as the biological indicators of diagnosis of epithelial ovarian cancer.2 TGF-β1 expression in ovarian cancer tissue has no relation with the age, histological type, histopathological grading and lymphatic metastasis of the patients and presents positive correlation with the clinical stages which shows that TGF-β1 contacts closely with the progress of ovarian cancer deterioration.3 The expression between TGF-β1 and MDR1 has a positive correlation in epithelial ovarian cancer, which points out the synergistic effect between them exists in the occurrence and expanding of tumor, The expression between TGF-β1 and PTEN has a negative correlation in epithelial ovarian cancer showing the mutual antagonism with each other. |
