| Objective: Through the experiment research of the expression of MMP-28 and the changing situation in human lumbar nucleus pulposus tissue, and to evaluate the process of MMP-28 role and function in lumbar intervertebral disc degenerative disease, and analyzes its relevance with the degeneration grade.Methods: All patients had preoperative lumbar MRI detection, according to the imaging data and intraoperative seen lumbar disc degeneration can be divided into:The normal group, the protrude group, the extrude group and sequester group.The experiment through the hematoxylin-eosinstaining method(hematoxylin – eosin staining HE) to observe normal intervertebral disc tissue and degeneration of intervertebral disc tissue morphological differences, through immunohistochemical technique(Immunohistochemistry IHC) observation of the location of MMP-28 in lumbar nucleus pulposus tissue, using Western blot detecting MMP-28 protein expression at each level of the degeneration of intervertebral disc. All samples are according to the selection of standard materials, and the taken tissue samples derived from fiber ring of lumbar nucleus pulposus, as far as possible to avoid and eliminate the rest intra-spinal canal material influence on specimens of nucleus pulposus tissue,to make the experiment more able to objectively reflect the expression and change situation of MMP- 28 in the nucleus pulposus, the result of the experiment data was more reliable and convincing. With statistically significant differences(P<0.05).Results: The hematoxylin-eosin staining results show that the normal group and the degeneration of cell number and form all have obvious difference.Immunohistochemical results show the location of MMP-28 is in cytoplasm of lumbar nucleus pulposus cells, it was all expressed in the normal group and degeneration group, but the number of degeneration group cell staining and degree of degeneration were obviously in the normal group. Western blot showed that MMP-28 expression in all levels of intervertebral disc tissue, and intervertebral disc degeneration level had a correlation, each grade all had difference with statistical significance(P<0.05).Conclusions: The process of lumbar nucleus pulposus tissue degeneration has participation of MMP-28, association with its expression, at the same time, MMP-28 with the organization for lumbar disc tissue degeneration has the same increased aggravating levels trend. |
