| ObjectiveTo prove whether a high-fat diet cause NAFLD though unbalanced intestinalmicroecologe,and synbiotics improved NAFLD through regulating intestinal floraand influence the expression of TLR4.MethodsUsed high-fat diets to make SD rats NAFLD model,gave the rats synbioticstreatment for two weeks at different time points of NAFLD respectively.liver tissue ofrat was using for HE staining to observe the histopathological change, in the formof enzyme ratio method to measure the triglycerides (TG), cholesterol (TC) of livertissue,and the level of triglycerides (TG), cholesterol (TC), high-density lipoprotein(HDL), low density lipoprotein (LDL),fasting glucose, fasting insulin, AST, ALT ofserum, used the method of ELISA and immunohistochemical method to determineexpression level of toll-like receptor4(TLR4) of each intestinal mucosa, and used16s rRNA fluorescence quantitative PCR to detected the main intestinal flora of rats.Results1.The weight,liver index, TG,TC,AST,LDL,,fasting glucose, fasting insulin of fatdiet group were significantly higher than the normal diet group(P<0.05),and thetime of autonomous activities,feed and water intake of fat diet group weresignificantly lower than the normal diet group(P<0.05). After gaven the ratssynbiotics treatment for two weeks at different time points of NAFLD,the level ofTG,TC,AST,LDL,,fasting glucose, fasting insulin of serum of rats feed with fat diet were significantly reduced(P<0.05),liver index was lower(P>0.05),the time ofautonomous activities,feed and water intake were significantly increased(P<0.05).2. The liver pathological histology observation results show that in the fourth weekof fat diet feeding,the fat deposition distributed in the liver cells;in the eighth week offat diet feeding,simple fat change showed up;in the twelfth week of fat dietfeeding,the liver showed non-alcoholic steatohepatitis chang. Gave the rats synbioticstreatment for two weeks at three time points above-mentioned,the liver fat depositionbarely changed,and not deteriorated.3. The ELISA and immunohistochemical expression of TLR4of fat diet group wasincreased gradually,and significantly higher than the level of normol diet group((P<0.05).After gaven the rats synbiotics treatment for two weeks,the level of TLR4wassignificantly decreased(P<0.05).4.The expression of bifidobacterium and lactobacillus of fat diet group weredecreased,and significantly lower than normal diet group since the second week offeeding with fat diet(P<0.05),the expression of enterococcus was significantlyincreased(P<0.05). After gaven the rats synbiotics treatment for two weeks,theexpression of bifidobacterium and lactobacillus of fat diet group were significantlyraised(P<0.05), the expression of enterococcus was significantly reduced(P<0.05).ConclusionThe intestinal flora imbalance was showed before liver pathological histologyobservation showed fat deposition in NAFLD rats model induced by high-fat diet,theintestinal flora imbalance was a key pathway of NAFLD development.Synbiotics wasable to regulate the intestinal flora imbalance caused by high-fat diet,reduce theexpression level of TLR4of intestinal mucosa,and adjust biochemistry criterion ofserum and living quality,consquently improve the NAFLD. |
