Objective: To observe effect of the chaipu decoction serum on expression ofEotaxin and STAT6in Rat Trachea Epithelial; to explore the molecularmechanism of chaipu decoction in asthma treatment.Methods:48SD rats were divided into normal control group,asthmagroup,chaipu decoction group.Prepared asthma serum,normal serum and chaipudecoction serum. RTE were divided into six groups:asthma group(Agroup),normal control group(B group),chaipu decoction low-dose group(Cgroup),chaipu decoction dose group(D group),chaipu decoction high-dose group(Egroup) and control group(F group).Immunocytochemistry and RT-PCR wererespectively used to detect the level of Eotaxin and STAT6protein and mRNA inthe RTE.Results: HE staining showed that:Normal control group without inflammatorycell infiltration around airway;There are a large number of eosinophils andlymphocytes infiltration around bronchus and blood vessels,Part of the airwayepithelium fell off,Bronchial smooth muscle layer and basement membranethickening,Airway wall thickening and airway luminal stenosis.According to immunocytochemistry protein semi-quantitative analysis,theprotein optical density values(OD) of Eotaxin and STAT6in group A aresignificantly than group F,while the value of B,C,D,E OD were between the twogroups.The Eotaxin OD value of each group are respectively:group A0.37±0.04,group B0.31±0.02,group C0.23±0.02,group D0.22±0.03,group E0.21±0.01,group F0.16±0.03.STAT6OD values of each group arerespectively:group A1.35±0.08,group B1.14±0.12,group C0.60±0.08,group D0.51±0.07,group E0.44±0.07,group F0.43±0.04.Compared group A with group F,the Eotaxin and STAT6OD value are significant different(P<0.05).Compared togroup A and F,the Eotaxin and STAT6OD value are significant different in groupB(P<0.05),group A compared with the group C,D,E,group C compared with thegroup E,the difference was significant(P<0.05).There were not significantdifference between group C and D(P>0.05),and group D and E(P>0.05).According to RT-PCR semi-quantitative analysis,the relatively gray-scalevalue of Eotaxin mRNA in group A are significantly than group F,while the valueof B,C,D,E were between the two groups.The Eotaxin mRNA relativelygray-scale value of each group are respectively:group A0.58±0.05,group B0.57±0.06,group C0.54±0.03,group D0.32±0.03,group E0.31±0.03,group F0.30±0.03.Compared group A with group F,the Eotaxin mRNA relativelygray-scale value are significant different(P<0.05).Group A compared with thegroup D,E,the difference was significant(P<0.05).There was no significantdifference between group D and E(P<0.05),and group A and C(P<0.05).According to RT-PCR semi-quantitative analysis,the relatively gray-scalevalue of STAT6mRNA in group A are significantly than group F,while the valueof B,C,D,E were between the two groups.The STAT6mRNA relatively gray-scalevalue of each group are respectively:group A1.32±0.06,group B1.21±0.08,groupC0.83±0.03,group D0.54±0.04,group E0.45±0.02,group F0.32±0.02.Comparedgroup A with group F,the STAT6mRNA relatively gray-scale value aresignificant different(P<0.05).Compared to group A and F,the STAT6relativelygray-scale value are significant different in group B(P<0.05),Group A comparedwith the group C, D,E,the difference was significant(P<0.05).There was nosignificant difference between group C and D,group D and E(P<0.05),and groupC and E(P<0.05).Conclusion:1. Asthma serum can induce RTE increased the expression ofEotaxin and STAT6;2. Chaipu decoction serum can inhibit the expression ofEotaxin and STAT6of RTE.
