Background and purpose Esophageal squamous cell cancer(ESCC) ranks fourth among cancer related death in our country. Metastasis is the primary cause of the poor prognosis. DDR1has been reported to be able to promote cell migration through binding to NM IIA, but the specific mechanism is not clear. In our previous research we found Tyr1408of NM IIA heavy chain was highly phosphorylated in ESCC through phosphorylation MS analysis and its phosphorylation was closely correlated with DDRl; the overexpression of DDR1in ESCC can promote cancer invasion and metastasis. Phosphorylation of NM IIA heavy chain can inhibit self-assemble and thus promote cell migration. Hence we anticipate that DDR1promote ESCC metastasis by binding and phosphorylation of NM IIA Tyr1408. In this study, we try to prove that DDR1is closely correlated with phosphorylation of NM IIA, we will understand the relationship between phosphorylation of NM IIA and ESCC invasion and metastasis. All these study will provide a new direction for in-depth study of the mechanism of esophageal cancer metastasis.Results1.In the first chapter, we collected10cases of esophageal squamous cancer and corresponding para-cancer tissues. We using Western blotting to verify the amount of protein DDR1and NM ILA expression in esophageal squamous cell carcinoma and para-cancer tissues.2.1n chapter two, we cultured and profiled esophageal squamous cancer cell lines (KYSE-510), we verified the amount of protein DDRI and NM IIA expression in KYSE-510by Western blot,and try to find the relationship between protein DDRI and NMIIA.Conclusions1.The expression of protein DDRI and NM IIA in esophageal squamous cell carcinoma was significantly higher than the adjacent tissues2.KYSE-510esophageal cancer cell lines expression protein DDRI and NMII A.3.Co-immunoprecipitation experiments show DDRI protein closely related with protein NM IIA.
