ObjectiveStudy on extracting technology of total iridoid and total flavonoids from theleaves of Syringa veutina Kom and flavonoids compounds in the leaves ofSyringa veutina Kom by capillary electrophoresis method. Study on Syringaveutina Kom leaves in the of effects of Streptozotocin-induced hypoglycemiceffect of type1diabetes mellitus rats and determine the effective part andeffective components from the pancreas in glucose metabolism, lipidmetabolism, antioxidant, pathomorphological study on its hypoglycemic effect.MethodsThrough ethanol extraction by cold soak and reflow, petroleum etherdegreasing parts by ethyl acetate extraction separation of total flavonoidextracts, butanol extraction of parts separated total iridoid extracts. UltravioletSpectrophotometric method determination of extraction of flavonoids and iridoidfrom content. With high performance capillary electrophoresis with diode arraydetection method for simultaneous separation and determination of rutin,astragalin, isoquercitrin in the leaves of different origin of Syringa veutina Kom.Through the establishment of STZ induction rat model of type1diabetesmellitus, aqueous extracts of Syringa veutina Kom, extracts of total flavonoidsand total iridoid blood glucose test. Detection of targets for weight loss, fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), low densitylipoprotein (LDL-C), high density lipoprotein (HDL-C), insulin (INS),malondialdehyde content (MDA) and superoxide dismutase(SOD).Conventional HE dyeing and immunohistochemical staining, under opticalmicroscope observation of morphological changes of pancreas pathology ofeach group.Results1、UV-Spectrophotometric determination of Syringa veutina Kom leaves inethanol extracts of total flavonoids and total iridoid compounds. Ethyl acetatecontent of flavonoids in extracts of54.33%, N-butanol content of iridoid inextracts of66.00%. Extracts of content over50%, identified as the effective partof Syringa veutina Kom leaves.2、HPCE-DAD simultaneous determination of approach of rutin, astragalin,isoquercitrin in the leaves of Syringa veutina Kom content. Gel electrophoresiscondition: no paint stains standard fused silica capillary column (40cmx50μ m,length30cm).35mmol/l of borax solution (pH9.25) for the electrophoresisbuffer, isolation voltage of25kV, capillary temperature of25°c, measuring206nm wavelength, pressure injection5kPa3s. Regression equations were:Y=0.1655X-0.2085(r=0.9996); Y=0.2290X+0.0668(r=0.9999);Y=0.2255X-0.5145(r=0.9994). Average content of rutin RSD2.0000%;Averagecontent of astragalin RSD1.2913%; Average content of isoquercitrin RSD1.5763%.3、Syringa veutina Kom leaves extracts could improve external signs, andmental status in diabetic rats, increased activity, weight gain and improved "overmore than three little" syndrome.4、Aqueous extracts of Syringa veutina Kom leaves and Effective part of thetwo were able to significantly reduce fasting blood glucose in diabetic rats, increased serum insulin concentration. Hypoglycemic effect of the extract oftotal flavonoids and total iridoid have a certain dose-effect relationship. Amongthem, the low-dose of flavonoids extract and high-dose effect of total iridoidextracts have more stronger, aqueous extract has weaker.5、Aqueous extracts of Syringa veutina Kom leaves and Effective part of thetwo were able to significantly reduce total cholesterol, triglycerides, low-densitylipoprotein levels, elevated high-density lipoprotein levels of diabetic rats,easing abnormality of lipid metabolism.6、Aqueous extracts of Syringa veutina Kom leaves and Effective part of thetwo were able to significantly improve body activity of SOD in diabetic rats,scavenging free radicals, inhibiting lipid peroxidation and improving diabeticsymptoms of abnormal body.7、HE staining showed that light microscopically dosing groups of islet cellstructure changes less than high blood sugar in model groups, and rather thanthe control group difference. Compared with the model groups of high bloodsugar, immunohistochemical results showed that expression of Insulin frommedication groups markedly elevated, and expression of Glucagon significantlyreduced. Pathological examination revealed that flavonoids and total iridoidextracts in the leaves of Syringa veutina Kom may reduce islet cells shrink indiabetic rat, and reduce the pathological damage of pancreas cells. It had aprotective effect on induced islet injury in diabetic model.8、Most effective parts of lowering blood sugar of the leaves of Syringaveutina Kom were total flavonoids and total iridoid. Hypoglycemic mechanismmay promote insulin secretion, inhibit glucagon secretion, and pancreatic β cellrepair.ConclusionsTotal flavonoids and total iridoid in ethanol extracts from the leaves of Syringa veutina Kom are content determination by ultravioletspectro-photometric.Method is simple, reliable results. HPCE-DADdetermination of rutin, astragalin, isoquercitrin in the leaves of Syringa veutinaKom content. Method is sensitive, accurate. Most effective parts of loweringblood sugar of the leaves of Syringa veutina Kom are total flavonoids and totaliridoid. The extract has significant hypoglycemic effect, to improve lipidmetabolism disorders, to increase the levels of serum insulin, to increase thecapacity of antioxidant, to enhance insulin secretion and inhibit glucagonsecretion. Repair islet β cells, and treatment and prevention of diabetes and itscomplications.
