Objective:Capparis spinosa L seeds is a medicine used in clinic in Xinjiang Uygur Medicine, which has a long history.There are a few studies on the chemical constituents, but the lack of its quality control standard. Quality standards of Capparis spinosa L seeds were recorded in the Uygur Medicine Zhi (1999edition), which was low.The character identification, microscopic identification and TLC identification of β-sitosterol as control, no inspection and content determination were not written. Capparis spinosa L seeds standards were not included in the Chinese Pharmacopoeia. In order to improve quality standards of Capparis spinosa L seeds, TLC and fingerprint, moisture, extract (including water extract and alcohol extract), total ash and acid insoluble ash limit test for heavy metals and arsenic salt, as well as the main component of content determination of Capparis spinosa L seeds were established on the basis of quality standards guiding principles in the Chinese Pharmacopoeia (2010edition), based on which Quality standard draft was written.Methods:1The method for the identification of rutin in Capparis spinosa L seeds was established by TLC. Sample treatment methods, systematic, silica gel thin-layer plate type and temperature had effects on the method for the identification of rutin.2The method for determination of Rutin gallic acid, stachydrine hydrochloride in Capparis spinosa L seeds was established by HPLC, its methodology was verified systematically.3According to Chinese Pharmacopoeia, water,extract ten batches at Capparis spinosa L seeds (including water extract and alcohol extract), total ash and acid insoluble ash, heavy metal and arsenic salt limited were checked.4Fingerprint of Capparis spinosa L seeds was established by HPLC, methodology of which was researched.Mathematical models of the similarity analysis, cluster analysis and principal component analysis were used to the main chemical composition at Capparis spinosa L seeds.Results:1Used silica gel GF254plate in the identification of TLC, developing agent for methyibenzene:ethyl acetate:formic acid(5:4:1), at a wavelength of254nm, room temperature, the sample of rutin was separated with other dots.2Content determination of gallic acid, rutin, stachydrine hydrochloride was detectd by HPLC from ten batches in Capparis spinosa L seeds, results of rutin content in5.69mg-g-1~9.82mg-g-1range,the content of gallic acid in1.08mg-g-1~6.53mg-g-1range, stachydrine hydrochloride in13.41mg·g-1-24.59mg·g-1.3HPLC fingerprint of Capparis spinosa L seeds was built, RSD of the precision, stability and repeatability was in line with the requirements of;Main chemical components in HPLC fingerprint feature information was extracted and expressed by3mathematical models for ten different regions.The results showed that the main components have similar characteristics. Conclusion:1In accordance with the Chinese Pharmacopoeia (2010edition) quality standard guidelines requirements, TLC, water and total ash content determination method, check the project were established at Capparis spinosa L seeds, and quality standard Draft was completed HPLC fingerprinting of main components was comleted in Capparis spinosa L seeds, which can be used to control the quality of Capparis spinosa L seeds effectively.The methodology could be met the requirements of.3Quality standards of Capparis spinosa L seeds established in this study can improve Capparis spinosa L seeds quality standard of the Uygur Medicine Zhi (1999edition) reference.
