The study was conducted to select the optimum condition on the method of ethanol-ultrasonic extraction and separation with macroporous resin for total flavonoid from Capparis spinosa L. Research on its intervention effect of anti-inflammatory, analgesic, immunology and adjuvant-induced arthris to the experimental animal model.The experiment was taken the fruit of wild Capparis spinosa L. in Turpan as the material. Optimize the condition for extaction and separation of total flavonoid with macroporous resin by orthogonal design. The mocroporous resins were used to separate the flavonoids. The result selected the best resin by static and was used to dynamic absorption-elution. Anti-inflammatory and analgesic effect of flavonoids in fruit of Capparis spinosa L.be researched with acetic acid writhing, heating plate pain, xylene induced ears swelling and toes swelling induced by egg white test in experimental animal models. For observe the regulation of immunoreaction in normal and low-immunoreaction mouse, the low-immunoreaction mice modles be used by administrate cyclophosphamide and compared to levamisole as the positive control. Adjuvant-induced arthris rat modles were established by inject Freund's complete adjuvant for study on intervention effect to rheumatoid arthritis of total flavonoid in Capparis spinosa L., which include to observe swell trend of rats'toe, pathological changes of arthrosis synovial membrane tissues, and the cytokines in blood serum, routine check-up of blood, detect the erythrocyte sedimentation rate and rheumatoid factor.The optimum separation condition showed that ethanol concentration was 70%, ultrasonic time was 60 minutes, solid-liquid ratio was 1:40 at the room temperature. The best resin of H103 was screened out to fit the besrt isolation condition. The content of total flavonoids be isolated was 63.98% by standard curve, and concentrate was 15.45 mg/ml. The result shows that the extract could reduce pain and inflammation. The immunity of mouses was improved by administrate the extract. In the experiment of adjuvant-induced arthris rats, the swells can be catabatic, synovial membrane tissues can be protected, hyperplasy was marked, IL-6 and TNF-αwas decressed in blood serum, leucocyte and erythrocyte sedimentation rate were reduced which indicate that the rheumatoid arthritis can be intervented by taken flavonoid in Capparis spinosa L. Treatmen showed a certain relationgship with dasage.
