| Xiaoai Jiedu Recipe (XJR) is traditional Chinese medicine compound preparation under the guidance of "cancer toxin" theory of Professor Zhou Zhongying, which consists of Hedyotis diffusa Willd, Akebia quinata (Thunb.) Decne, Cremastra appendiculata (D.Don) Makino, Ophiopogonis Radix et al.Xiaoai Jiedu Recipe (XJR) has a solid fundamental clinical efficacy effect of traditional Chinese medicine and research application prospect. Presently, there are some reports about the reseach that XJR is pharmacological effects and clinical application at home and abroad, chemical research and material basis has not been reported.The chemical compostion of Chinese medicine compound preparation will play an important material basis when it applied in the clinical treatment. Due to having rarely rules of mass spectrometry data, qualitative research of the mass spectrum in many medicinal plants is almost blank. Thence, to study on single herbs chemical composition of XJR and summarize the mass spectrometry cracking rule of main compounds will lay the foundation for identification of Chemical composition of XJR.Using UFLC-Q-TOF/MS technology, the chemical composition of the single herbs and XJR were identified. XJR was divided into the part of petroleum ether, dichloromethane, ethyl acetate, n-butanol by the method of solvent system. By rat model of liver cancer Walker-256, antitumor effective parts were preliminary screened out, which were methylene chloride and n-butanol part. Then, chemical compositions of the effective parts by UFLC-Q-TOF/MS technology were analyzed. The XJR Effective Substances of Anti-tumor by this research was primarily illustrated in this thesis. Forty-six triterpenoid saponins from Akebia quinata (Thunb.) Decne were identified. Thirty-five homoisoflavonoids and thirty-six steroidal saponins in Ophiopogon were detected.fifteen compounds in cremastra appendiculata (D.Don) Makino were identified mainly for benzyl and dihydrogen class. seventy-eight chemical components could been identified mainly from water extract of XJR and ninty chemical components could been identified mainly from ethanol extract of XJR.The Chapter three, to set up a chemical composition database of XJRThe chemical composition of corresponding database on the basis of our investigations and literature research were established.The Chapter four, Medicinal material basic research of XJRUsing system solvent separation method, Xiaoai jiedu Decoction will be separated of petroleum ether part, methylene chloride part,ethyl acetate, n-butyl alcohol part and water area. The Walker-256in the rat model of liver cancer was adopted to screen cancer the effective parts of XJR. By compared with model group effective, the anti-tumor rate of n-butyl alcohol part and methylene chloride part were36.96%and25.07%respectively. Then, the chemical composition structure of n-butyl alcohol part and methylene chloride part were analyzed by means of UFLC-Q-TOF/MS method. Forty-eight chemical components could been identified mainly from methylene chloride part of XJR and twenty-six chemical components could been identified mainly from ethyl acetate part of XJR. Sixty-eight chemical compositions from n-butyl alcohol part were identified, which mainly belongs to flavonoids and saponins.Xiaoai Jiedu Recipe (XJR) was systematically studied based on UFLC-Q-TOF/MS method, which lay a foundation of the further analysis of the chemical compositions and effective substances. |
