ObjectiveImmune thrombocytopenia (ITP) is immune disease.Interleukin-27(IL-27) canregulate T cell differentiation. However, it is unclear whether IL-27correlates withthe dysfunctions of T cell differentiation in ITP patients. Thus,in order to determinethe roles of IL-27in ITP, we studied the expression of IL-27, the percentage of Th1,Th17cell in ITP patients.MethodsThirty-six ITP patients were enrolled in this study, from the First AffiliatedHospital of Zhengzhou University, Department of Pediatrics. The diagnosis of ITPwas based on American Society of Hematology (ASH)2011evidence-baseddiagnostic criteria. The normal control group consisted of20clildren healthy controls.Patients with active or chronic infections, malignant hematological system diseases,tumor or connective tissue diseases were excluded.Th17and Th1cells in both ITP patients and controls were examined by flowcytometry. Plasma interleukin-27(IL-27) level was measured by enzyme linkedimmunosorbent assay (ELISA).ResultsCompared with the healthy controls,the percentage of Th17cells wassignificantly higher in ITP patients[(0.77±0.25)%vs (0.42±0.20)%), P﹤0.001]; The percentage of Th1cells was significantly lower in ITP patients([3.27±1.02)%vs(5.69±1.60)%, P﹤0.001];We also observed a statistically significant drecrease inthe plasma IL-27levels of ITP patients[(177.45±68.70)ng/L vs(309.07±138.90)ng/L, P﹤0.001].In the ITP patients,we also observed a positive correlation was found betweenthe percentage of Th1cells and the plasma IL-27concentration (r=0.629,P﹤0.001);a negative correlation was found between the percentage of Th17cells and the plasmaIL-27concentration(r=-0.548,P=0.001).ConclusionIn ITP children, abnormalities were found in the ratio of Th1and Th17cells andthe level of IL-27. The level of IL-27was correlated with the ratio of Th1and Th17cells. IL-27might play a role in the pathogenesis of ITP by Involving in Tlymphocyte subsets disorder. |
