Objective:By establishing the rat model with chronic fatigue, we observe GuiluYishen formula particles on chronic fatigue rat general situation and behaviouralindicators (exhaustion swimming time、 across the grid and standing movementtimes),observe the effect of Guilu Yishen formula particles on the contents of COXand Cyt of chronic fatigue rat skeletal muscle mitochondria,in order to explore theanti-fatiguemechanism of Guilu Yishen formula particles to treatment CFS.Methods:1.Modeling、Grouping:40SPF SD male rats,weight200±20gram, afteradaptive feeding1week,we randomly selected16without modeling and theremaining24carried modeling.we adopted forced exhaustion swimming,bound andclip tail composite factors stimulation to construct the chronic fatigue ratmodel.After the the success of modeling,the modeling was randomly divided into3groups which group has8rats,respectively, the model group, the Desert cistanchekidney group and the Guilu Yishen group; without modeling was randomly dividedinto two groups which group has8rats, named the normal group and the Guilucontrol group.2.Method of administration:From the next day after the end of themodeling,the normal group and the model group were gave saline gavage,the Guilucontrol group and the Guilu Yishen group were gave Guilu Yishen formula particlessuspension liquid gavage,the Desert cistanche kidney group was gave the Desertcistanche kidney particle suspension liquid gavage.Rats in each group wereadministered1times/d,for14consecutive days.3.Extract tissue:we took the piece ofbilateral quadriceps(1g)of rat in each group,made into homogenising fluid,gotmitochondria by differential centrifugation.useing the ultrasonic cell crushingapparatus damaged the mitochondrial membrane and saving them at lowtemperature.The whole operation must be on ice.4.The detecting of experimentalindicators:We measured the rat body weight、 general situation and behaviouralindicators (exhaustion swimming time、across the grid and standing movement times)in the day before modeling,after modeling and after the last administration.We test the contents of COX and Cyt in difference spectroscopy by using ultravioletspectrophotometer.We collected the datas and used SPSS18.0statistical software toprocess them.Results:1.General Situation:a.The rat’s change of eating、 faces and fur:Beforemodeling,there is no difference with eating、faces and fur between each group ofrats.After modeling,every modeled group rats had different degrees of "Spleen"Symptoms:reduced food intake and water consumption,grew slowly,squinting lazymove,loose stools,hair lost luster;the case of the normal group and the Guilu controlgroup as usual.After gavage,the food intake and activity of the Desert cistanchekidney group’s and Guilu Yishen group’s rats were increased significantly,inaddition,the fur became more glossy.The Guilu Yishen group had more pronouncedincreasing and the change of model group was not significantly.b.Changes of rat’sweight:Before modeling,every goup’s weight had no difference.After modeling,compared with the normal group,the weight of the model goup、Guilu control groupand Desert cistanche kidney group reduced significantly which had significantdifference(P<0.05);there was no difference between them(P>0.05).After gavage,themodel group rats’ weight losser than the other4groups,which had significantdifference(P<0.05).The weight of normal group was higher than Desert cistanchekidney group and had significant difference(P<0.05).Compared with the normalgroup,the weight of the Guilu Yishen group has no significant difference(P>0.05).2.Behavioural Indicators:before modeling,each group rats’s exhaustion swimmingtime and the across the grid and standing movement times had no significantdifference (P>0.05).After modeling the end, Compared with the normal group,thethree model group had significant difference (P<0.05) and there had no significantdifference between the three model group.After gavage,the time and movements ofthe normal group were less than the Guilu control group,which had significantdifference (P<0.05).There was no difference between the normal group、the Desertcistanche kidney group and the Guilu Yishen group,which had no significantdifference (P>0.05).The model group rats’ exhaustion swimming time and the acrossthe grid and standing movement times were less than the other4groups,which hadsignificant difference(P<0.05).Compared with the Desert cistanche kidney group,theGuilu Yishen group’shad significant difference (P<0.05).3.The content’s detection of COX and Cyt:Compared with the normal group,thecontent of COX and Cyt of Skeletal muscle’s mitochondria on the Guilu Yishen group’s had no difference,and there was no significant difference(P>0.05);Themodel group rats’ contents of COX and Cyt were less than the other4groups,whichhad significant difference(P<0.05).The contents of the Guilu Yishen group washigher than the Desert cistanche kidney group,which had significantdifference(P<0.05).The contents of the normal group was higher than the Desertcistanche kidney group and which one was less than the Guilu control group andboth of them had significant difference (P<0.05).Conclusion:1.In this experiment,we take a composite way to build the rat modelwith chronic fatigue:physical fatigue which caused by excessive movementcombined with psychological stress.That way highly simulates the human body’sinternal and external environment which caused the chronic fatigue syndrome toensure the rationality and science of the animal model,so that the results moreaccurate.2.Guilu Yishen formula particles can obviously extends the chronic fatigue ratweight、swimming exhaustion time,shortens the across grid and standing movementtimes and also can improve their mental and physical fatigue.The medicinal effectsbetter than the Desert cistanche kidney group.In addition,it can improve the fatigueresistance of normal rats and have the potential efficacy of “Preventive treatment ofdisease”.This result provides a basic research data for the clinicalapplication andpromotion of Guilu Yishen herbal extract granules.3.Guilu Yishen formula particles can obviously improve the content of COX and Cytof Skeletal muscle’s mitochondria on chronic fatigue rat and corrective the smstatusof mitochondrial metabolism which is better the Desert cistanche kidney group.Theexperimental results prove the scientific and practical of treating CFS by GuiluYishen formula particles through the microscopic point of view and supplement theanti-fatiguemechanism of Guilu Yishen formula particles.
