| ObjectiveThe hyperlipaedemia model using male C57BL/6mice fed high-fat diet was adopted to explore the mechanism of traditional Chinese classic herbal formula Huanglianjiedu decoction impacting on inflammatory factors in hyperlipidemic mice. And using PCR-DGGE fingerprinting and quantitative real-time polymerase chain reaction technology researched the impact of Huanglianjiedu decoction on the characteristic of intestinal flora in hyperlipidemic mice.MethodsForty-eight Male C57BL/6mice were randomly divided into six groups: normal control group, high-fat control group, normal diet plus antibiotic group, High fat diet plus antibiotic group, low dose Huanglianjiedu decoction group and high dose Huanglianjiedu decoction group. The mice fed high-fat diets for6weeks were administered with Huanglianjiedu decoction at the doses of195,390mg/mL to create high and low dose groups. Antibiotic group fed either conventional diet or high fat diet for6weeks, then drank Antibiotic freely for2weeks. After8weeks, the body weight, and serum parameters such as TC, endotoxin, TNF-α and IL-6were determined. Then, the effect of high-fat diet on the intestinal flora in mice were investigated by PCR-DGGE and real-time fluorescent quantitative PCR technology.ResultsThe blood total cholesterol(TC) levels of high-fat control group were significantly increased. The concentations of endotoxin, tumor necrosis factor(TNF-α) and interleukin-6(IL-6) show significantly increased in the high-fat control group, and low dose Huanglianjiedu decoction group were lower than in the high-fat control group. The diversity index and species abundance of the intestinal flora in the high-fat control group are higher than other groups. Compared with the high-fat control group, the expression of bacteriodes in the normal control group and the high dose Huanglianjiedu decoction group are significantly increased;the expression of Bifidobacterium in the normal control group and the low dose Huanglianjiedu decoction group are significantly increased. Compared with the high-fat control group and the normal control group, the expression of Lactic Acid Bacteria in both of the two Huanglianjiedu decoction group are significantly increased.ConclusionsLow dose Huanglianjiedu decoction could significantly reduce serum endotoxin, TNF-α and IL-6content. The community of the intestinal flora could be changed by high-fat diet. The probiotics in gut could be inhibited in hyperlipidemiamice, but the low dose Huanglian jiedu decoction play a important role in the growth of the probiotics. |
