BackgroundBreast cancer is one of the three common malignant tumors in women, andthe incidence of this disease has risen to number one in female malignant tumors.Especially in recent years,the incidence rate and mortality rate of breast cannerhave been accelerating worldwide.As we are not clear about the pathogeniccauses and mechanism of breast canner molecules’ biological base,and the earlydetection and prognosis are not perfect,so the study of breast canner’s genebiology and molecular biology targeted treatment have become a new direction.We can hence provide an effective treatment scheme of molecular biology forthe sick women’s later years and recurrence of the disease.It is very importantfor an early diagnosis and treatment of breast canner,so early diagnosis andtreatment from gene level has become a hot research topic in many biginstitutions.ObjectivesThrough checking the expression differences of insulin-like growthfactor-1(IGF-Ⅰ)and Ki-67in the breast cancer tissues, breast hyperplasia tissusand normal breast tissue,the author explores the expressions and clinicalsignificance of IGF-Ⅰ and Ki-67in breast cancer.Method1. Detecting the expressions of IGF-Ⅰ and Ki-67in70cases of the breastcancer tissues, breast hyperplasia tissus and normal breast tissue by means ofimmunohistochemistry (SP);defining their accurate expression location,andmaking a statistical analysis of the expression rate of positive cells.2.Detecting the contents of IGF-Ⅰ in70cases of the breast cancer tissues,breast hyperplasia tissus and normal breast tissue with reverse transcribing polymerase chain reaction (RT-PCR) technique, and making a semi-quantitativeanalysis of the expression difference between them.3. The expressions of IGF-Ⅰ and Ki-67are examined by RT-PCR andimmunohistochemistry (SP).And the expression differences were analyzed bySPS13.0。Results1. The levels of IGF-Ⅰ increased from the normal breast tissue, breasthyperplasia tissus to breast cancer tissues by means of immunohistochemistry(SP); The mRNA levels of IGF-Ⅰ inc Sreased from the normal breast tissue,breast hyperplasia tissus to breast cancer tissues,The ratios of IGF-Ⅰ/β-actinwere0.169±0.028、0.339±0.043、0.621±0.035,respectively,and the differencebetween any2groups was significant(P <0.05).2. IGF-Ⅰ was closely correlated with lymphatic metastasis andpathological stages,statuses of ER of breast cancer(P<0.05),while insignificantly correlated with pathological types (P>0.05).3.Ki-67was closely correlated with lymphatic metastasis and pathologicalstages,statuses of ER of breast cancer(P<0.05),while insignificantly correlatedwith pathological types (P>0.05).4. The expressions of IGF-Ⅰ and Ki-67were Consistent in breast cancertissues.Conclusions1.IGF-Ⅰ and Ki-67are closely related to the local infiltration and distantmetastasis of breast cancer; The high expressions of IGF-Ⅰ and Ki-67mayindicate a higher degree of malignant tumor or a poor prognosis.2. The high expression of IGF-Ⅰ and Ki-67may be one of the pathogenesisof breast cancer.The abnormal protein expressions of IGF-Ⅰ and Ki-67showthat they are both involved in the occurrence,development and metastasis ofbreast cancer.The expressions of IGF-Ⅰ and Ki-67are positively correlated withtumor invasion,IGF-Ⅰ and Ki-67can be used as an independent molecularmarker for determining the prognosis and planning the postoperativetreatment.The expressions of IGF-Ⅰ and Ki-67protein are associated withlymph node metastasis in breast cancer, IGF-Ⅰ and Ki-67may become an important prognostic marker for breast cancer.For the treatment of metastaticpatients,the evaluation of IGF-Ⅰ、 Ki-67status should be performed inneoplastic tissue from metastatic site,when-ever possible.
