| Objective:Before and after periodontal non-surgical treatment of chronic periodontitis, we will study the change of elastase(EA) and prostaglandin E2(PGE2) in gingival crevicular fluid and if minocycline hydrochloride will influence that change.Method:We select25adult periodontitis according to the standard that everyone has teeth with more than5points with no less than3mm depth of periodontal pocket and no less than1mm depth of attachment loss in left and right of mouth. According to the standard, we choose3points to do periodontal clinical examination which include probing depth(PD), attachment loss depth(AL), gingival index(GI), then aetract same amount of gingival crevicular fluid with every filter paper in every point and record the time of extracting(GCFtime). Then, put the filters into Eppendorf(EP) shocking for one hour with oscillator and centrifuging for five minutes with centrifugal machine, then extract supernate into new EP and storage in a refrigerator of seventy degrees below zero. After extracting gingival crevicular fluid, take periodontal non-surgical treatment to every periodontitis that include ultrasonic tooth with ultrasonic dental unit, scaling and root planning, then inject minocycline hydrochloride into the right points every week for four times. Six weeks after periodontal non-surgical treatment, do the same periodontal clinical examination and gingival crevicular fluid extracting as before. Then,take out all the gingival crevicular fluid sample warming to normal temperature, and measure the concentration of PGE2and EA with enzymelinked immunoserbent assy. Analyze all the data with spss13.0. Result:PD,AL,GI all reduce after periodontal non-surgical treatment with statistical significance(P<0.01); GCFtime increase after periodontal non-surgical treatment with statistical significance(P<0.01); The concentration of PGE2and EA all reduce after periodontal non-surgical treatment with statistical significance(P<0.01); The change of PD, AL, GI, FCFtime, PGE2, EA between the left points with no minocycline hydrochloride and the right points with minocycline hydrochloride has no statistical significance(P>0.05).Conclusion:For chronic periodontitis, the concentration of PGE2and EA in gingival crevicular fluid can be reduced by periodontal non-surgical treatment and it could be referencr standard of periodontal disease severity and periodontal non-surgical treatment effect. Minocycline hydrochloride affect little on periodontal clinical index and the concentration of PGE2and EA in gingival crevicular fluid, it plays an assistant role in the treatment of periodontitis. |
