Objective1.To detect the effect between X-ray irradiation and EMT2.To exam the change of key molecular which associate with epithelial-mesenchymal transition in colorectal SW480carcinoma cells. Explore the may signaling pathway of X-ray induced epithelial-mesenchymal transition.Methods1.The human colorectal carcinoma cells SW480were divided into five groups,and X-ray irradiation would irradiate these cells with0Gy、2Gy、4Gy、6Gy and8Gy respectively. After irradiated, cells were cultured48h before experiment.2.We detected the expression of Vimentin by using Western-blot.3.Using qRT-PCR and Western-blot detect the expression of K-ras and Smad3. Analysis the perhaps signaling pathway of the X-ray-induced EMT.Results1.After X-ray irradiation, the expression of Vimentin in2Gy、4Gy、6Gy and8Gy were significantly increased compared with non-irradiated group.2.QRT-PCR showed that the expression of K-ras was decreased in4,6,8Gy groups compared with0Gy group(P<0.05). There is no significant changes between between0Gy and2Gy group(P>0.05).3.Using QRT-PCR detect the expression of Smad3, and we found that Smad3was increased significantly in2Gy、4Gy、6Gy and8Gy groups(P<0.05).There were no significantly difference among experiment groups(P>0.05).4.We detected the expression of K-ras using Western-blot, found that, the K-ras was decreased in2Gy、4Gy、6Gy and8Gy groups(P<0.05)significantly compared with0Gy group. 5.We also using Western-blot to detect the expression of Smad3, found that, the Smad3was increased significantly in4Gy、6Gy and8Gy groups(P<0.05), but there is no significantly difference between0Gy and2Gy groups(P>0.05).Conclusion1.X-ray irradiation can induce epithelial-mesenchymal transition in human colorectal cancer cells.2.X-ray may through Smads signaling pathway to induce EMT in human colorectal cancer cells.3.There is no close association between X-ray-induced EMT and K-ras.
