With the increase of morbidity in ischemic stroke (IS) every year, the huge costof treatment has become a vital economic, health and social problem. Therefore,early prevention of ischemic stroke is important. The aging problem is serious inLiwan District of Guangzhou. In this case-control study of the local old female, it isused to find the association of numerous factors and IS.With Human Genome Project completed, scientists focus on heredity tomanifest the pathogenesis of ischemic stroke. X chromosome inactivation is asignificant part in human epigentics research, which is related to various diseases.On the basis of studies in ischemic stroke in the world, the principle and mechanismof X chromosome inactivation has been added to our study. It is the first time to findthe relationship between female ischemic stroke and X chromosome inactivation,comparing the differences in patients with ischemic stroke and healthy female. It isbeneficial for women to prevent ischemic stroke and screen high-risk individuals.[Objective]1. To investigate the association between traditional influencing factors and ISin local old female.2. To investigate the possible association between X chromosome inactivationand IS in local old female.[Method]1. A case-control study was used to determine the possible association betweentraditional factors and IS in local female. It was performed that150cases were thoseonset IS patients registered and150controls were selected with the approximate age. The cases and controls were interviewed with the same questionnaire, and the bloodsamples were drawn in terms of the same conditions and standards. Univariate testand multiple logistic regression models were ultilized to analyze the above data.2. DNA of30IS cases and30controls in female was amplified to analyze thepolymorphism patterns of the AR gene. Only the DNA of heterozygous was bisulfitetreated and performed with a methylation-specific PCR. Then the polymorphismpatterns of products were analyzed again.[Result]1. Univariate logistic regression demonstrated that risk factors of local femaleIS included overweight, smoking, hypertension, diabetes mellitus anddyslipidemia.,while moderate physical training was inversely associated with IS. Themultiple logistic regression models proved that hypertension, diabetes mellitus, TGlevels and family history were positive correlation factors, however, physicaltraining was a negative one.2. The ratio of AR gene homozygotes in IS was similar to of normal controls.With chi-square test, the SXCI showed the greatest statistical significance in itsassociation with ischemic stroke.[Conclusion]1. The traditional risk factors are still main reasons of patients with IS in thelocal female population. Therefore it is an important measure to prevent IS incommunity population to propose healthy life style, control blood pressure, glucoseand fat. Moreover, to investegate family history is beneficial for the prognosis of IS.2. SXCI is highly correlated with IS, which provides the epigentic evidence inthe etiology of female IS.
