Objective1.To detect the effects of X-ray irradiation on the epithelial to mesenchymal transitionin human colorectal carcinoma SW480cells which cultured in vitro.2.To observe the influence of X-ray irradiation on the biological characteristics inX-ray irradiated SW480colorectal carcinoma cells. And we also observe the capacityof invasion and migration after SW480carcinoma cells were irradiated.Methods1.The human colorectal carcinoma SW480cells were divided into five groups,andX-ray irradiation would irradiate these cells with0Gy、2Gy、4Gy、6Gy and8Gyrespectively. After irradiated, cells were cultured48h before experiment.2.The cell morphology was observed after48h of X-ray irradiation.3.We detected the changes of0Gy,4Gy and8Gy groups cell invasion and migrationcapacities after X-ray irradiated24h by using Transwell.4.Using QRT-PCR and Western-blot to detect the expression of epithelial targetsE-cadherin and mesenchymal maker Vimentin. Analysis the relationship betweenX-ray and EMT in human colorectal carcinoma SW480cells.Results1.After X-ray irradiation, the cell morphology was changed in2Gy,4Gy,6Gy and8Gy groups, which evidenced by intercellular separation, loss of polarity, spindle-cellshape and pseudopodia formation. These changes were consistent with EMT. 2.The X-ray irradiation increased cell invasion significantly in4Gy group(P<0.05)and in8Gy group(P<0.05) compare with the control0Gy group, respectively. And, theinvasion of8Gy was increased compared with4Gy group(P<0.05).3.The X-ray irradiation also enhanced the cell migration significantly in4Gy(P<0.05)and in8Gy(P<0.01) compared with the0Gy, respectively. And, there is nosignificantly difference between4Gy and8Gy groups.4.QRT-PCR and Western-blot showed that the expression of E-cadherin wasdecreased in2Gy,4Gy,6Gy and8Gy groups compared with0Gy group(P<0.05), andthe higher dose the more it decreased.5.We also detected the expression of Vimentin using QRT-PCR and Western-blot,found that, after X-ray irradiation the expression of Vimentin was enhancedsignificantly in2Gy,4Gy,6Gy and8Gy groups after X-ray irradiation compared with0Gy group(P<0.05), and the more X-ray dose the more it enhanced.Conclusion1.X-ray irradiation can induce EMT in human colorectal carcinoma cells.2.There may be a positive correlation between X-ray dose and EMT.3.X-ray irradiation can enhance human colorectal carcinoma cells invasion andmigration capacities.
