Objective: To explore the effect of Echinococcus multilocularis vesicle fluid(EmF) on mitogen activated protein kinase(MAPK)in th hepatic stellate cell of rats.Method: The rat hepatic stellate cells HSC-T6were cultured with Different proteinconcentration of EmF, collecting cells after24h, collecting control cells (withoutintervention of EmF) at the same time. To detect the change of ERK1/2、JNK1/2andp38MAPK with Florescent real-time quantitative (RT-PCR).Result: Most cells in EmF are with pyknotic and show fall off a precursor;afterbeing cultured with HSC for24h. some cells of1/2The original fluid groupconclusion are with pyknotic showing fall off a precursor,some cell of HSC-Tcontracting spindle growth, bring forth many slender pseudopodia cells,as large astwice size of normal condition.Some cells of1/4The original fluid group contractingspindle growth, bring forth many slender pseudopodia,but most cells showing asadhesive, large-body irregular polygon, connect with other cells into flake withShorter pseudopodia.Less than1/8The original fluid group show no difference withcontrol cells. Real time PCR shows sthat ERK1/2、JNK1/2and p38MAPK mRNAsignificantly increase when the concentration of EmF>0.4mg/mL, beingoverexpression compared with the control group at the Concentration of6.8mg/mL,and there was significant difference comparing with contrastgroup(P<0.05).Conclusion: Various concentrations EmF can activate the MAPK pathway of therat hepatic stellate cell by integration with the rat hepatic stellate cell surface receptor.The most significant occurs at the concentration of6.8mg/mL.
